The effects of prostaglandin F2α (PGF2α) on intracellular Ca2+ concentration ([Ca2+]i) and inositol phosphate (IP) generation in human myometrial cells were evaluated and compared to the effects of oxytocin. Basal [Ca2+]i levels were 146 and 153 nM in the absence and presence of 1 mm extracellular Ca, respectively. In Ca-containing medium, both PGF2α and oxytocin significantly (P < 0.01) increased [Ca2+]i over control values, eliciting half-maximal stimulation (ED50) at 4 and 1 nm, respectively. In Ca-free ...

The specific binding of prostaglandins (PG) F2 alpha and E2 was studied in a rat myometrial membrane-enriched fraction during the latter part of gestation and parturition, as well as in the postpartal period. Tritiated PGE2 and PGF2 alpha binding was specific, saturable, time dependent, and directly proportional to the amount of membrane protein. Scatchard analysis indicated the presence of high-affinity (Kd2) and low-affinity (Kd2) binding sites for both PGs. The affinity of both binding sites for ...

Digitonin-permeabilized avian granulosa cells were used as a model to study the effects of LH, 8-bromo-cAMP (8-Br-cAMP), and forskolin on intracellular calcium mobilization. LH caused a biphasic calcium efflux. There was a small but significant (P < 0.05) calcium release within 1–2 min (rapid phase) with an ED50 of 35 μg/ml, followed by a larger one after 5 min (slow phase) with an ED50 of 10 ng/ml. The intracellular calcium antagonist TMB-8 inhibited the action of LH in both phases. Forskolin had no ...

Previous studies in this laboratory have shown a remarkable increase in the capacity of avian granulosa cells to produce progesterone during the last 2-3 days of folliculogenesis, with concomitant increases in the activities of key steroidogenic enzymes. In view of the proposed involvement of inositol phospholipids and their hydrolytic products in signal transduction in steroidogenic cells, we investigated in this study the influence of follicular maturation on phosphoinositide-specific phospholipase ...

In this article we study the multicast routing problem in all-optical WDM networks under the spare light splitting constraint. To implement a multicast session, several light-trees may have to be used due to the limited fanouts of network nodes. Although many multicast routing algorithms have been proposed in order to reduce the total number of wavelength channels used (total cost) for a multicast session, the maximum number of wavelengths required in one fiber link (link stress) and the end-to-end ...

Human serum paraoxonase-1 (PON1) protects lipoproteins against oxidation by hydrolyzing lipid peroxides in oxidized low-density lipoprotein (LDL); therefore, it may protect against atherosclerosis. Changes in the ratio of high-density lipoprotein (HDL) subfractions may alter the stability and the antioxidant capacity of PON1. The aim of the study was to examine the effect of atorvastatin treatment on the distribution of HDL subfractions, LDL size, cholesteryl ester transfer protein (CETP), lecithin- ...

Single blastocyst transfer is regarded as an efficient way to achieve high pregnancy rates and to avoid multiple pregnancies. Risk of cancellation of transfer due to a lack of available embryos may be reduced by early prediction of blastocyst development. Time-lapse investigation of mouse embryos shows that the time of the first and second cleavage (to the 2- and 3-cell stages, respectively) has a strong predictive value for further development in vitro, while cleavage from the 3-cell to the 4-cell ...

The primary aim of this study was to evaluate the effects of arachidonic acid (AA) on calcium mobilization from intracellular compartments in digitonin-permeabilized granulosa cells isolated from the largest preovulatory follicles of laying hens. At low concentrations (ED50 0.2 μM) AA released 35% 45Ca from the endoplasmic reticulum (ER), whereas at higher concentrations (ED50 16 μM) it stimulated 45Ca efflux from mitochondria. These effects of AA were mimicked at 10–20 times lower concentration by ...

This book offers a comprehensive thousand-year history of the land, people, society, culture and economy of Hungary, from its nebulous origins in the Ural Mountains to the elections of 1988. It tells above all the thrilling story of a people which became a great power in the region and then fought against - and was invaded by - Ottomans, Germans and Soviets. The Hungarian people preserved nevertheless a continuous individuality through its Ural-born language and a specifically Hungaro-European culture. ...

Vitrification of in vitro (IVP)-produced bovine blastocysts is well established, reaching post-warming hatching rates close to 70–80% in vitro. Nevertheless, improvements still are needed regarding realizable pregnancy rates. High hydrostatic pressure (HHP) treatment of fresh boar semen before freezing increased the litter size achieved by insemination of frozen–thawed boar semen (Kuo et al. 2007 6th Int. Conf. Boar Semen Pres, Alliston, ON, Canada, poster #22); HHP treatment-related improvements were ...

The present paper describes a method which uses high hydrostatic pressure as a pre-treatment to in vitro matured porcine oocytes to improve their survival rates in the subsequent processes including cryopreservation, parthenogenetic activation and embryo culture. In Experiment I oocytes were treated with different pressure impulses in the range of 20–80 MPa (200–800 times greater than atmospheric pressure) for 30–120 min at 24 °C. For parthenogenetic activation a single dc of 12.5 kV/cm was used, ...

Network survivability is becoming an important issue and a topical subject in WDM optical mesh networks. Many works have studied network survivability. However, few works have focused on survivability in multi-domain optical networks. This paper reviews the literature on survivability against failures in multi-domain optical networks. The main objective of this study is to evaluate and analyze existing solutions and to compare their performance in terms of different criteria: resource utilization, ...

Redundant sensing capabilities are often required in sensor network applications due to various reasons, e.g. robustness, fault tolerance, or increased accuracy. At the same time high sensor redundancy offers the possibility of increasing network lifetime by scheduling sleep intervals for some sensors and still providing continuous service with help of the remaining active sensors. In this paper centralized and distributed algorithms are proposed to solve the k-coverage sensing problem and maximize ...

The construction of light-trees is one of the principal subproblems for all-optical multicast routing in sparse splitting wavelength division multiplexing (WDM) networks. Due to the light splitting constraint and the absence of wavelength converters, several light-trees may be required to establish a multicast session. However, the computation of the cost-optimal multicast light-trees is NP-hard. In this paper, first we study the cost bounds of the light-trees built for a multicast session in unweighted ...

Previous findings suggest a potential therapeutic action of relaxin, the putative vasodilatory signal of normal pregnancy, in some forms of cardiovascular disease. However, the mechanisms underlying the beneficial effects of relaxin have not been fully elucidated. The purpose of this study was to determine whether the vasodilatory effects of relaxin are dependent on activation of NO synthase. We examined the effect of relaxin in male Sprague-Dawley rats given angiotensin II (Ang II; 200 ng/kg per ...

In this paper, we define the cost optimal solution of the multi-constrained multicast routing problem. This problem consists in finding a multicast structure that spans a source node and a set of destinations with respect to a set of constraints, while minimizing a cost function. This optimization is particularly interesting for multicast network communications that require Quality of Service (QoS) guarantees. Finding such a structure that satisfies the set of constraints is an NP-hard problem. To ...

The role of Mg2+ in the binding of ADP and ATP to pig muscle and yeast 3-phosphoglycerate kinases has been studied by equilibrium dialysis. Whereas the Kd of ATP binding varies between 0.17 and 0.23 mM (S.E.M. 0.03 mM) for both enzymes, independently of the presence of Mg2+, the Kd values for ADP and MgADP binding are in the range 0.18-0.27 mM (S.E.M. 0.04 mM) and 0.05-0.06 mM (S.E.M. 0.01 mM) respectively. Thus Mg2+ exclusively tightens the interaction of ADP, but not of ATP, with the protein molecule. ...
