The roles played by the endogenous angiogenesis inhibitor thrombospondin-1 (TSP-1) in the early stages of multi-step carcinogenesis and in the control of hematogenous versus lymphatic metastasis are unknown. To investigate these issues we compared tumor development in normal mice and in transgenic mice with targeted overexpression of TSP-1 in the epidermis following a standard two-step chemical skin carcinogenesis regimen. Overexpression of TSP-1 resulted in delayed and reduced development of premalignant ...

Trehalose is a non-reducing disaccharide that is used as an osmolyte, transport sugar, carbon reserve and stress protectant in a wide range of organisms. In plants, trehalose 6-phosphate (Tre6P), the intermediate of trehalose biosynthesis, is thought to be a signal of sucrose status. Trehalose itself may play a role in pathogenic and symbiotic plant-microbe interactions, in responses to abiotic stress and in developmental signalling, but its precise functions are unknown. A major obstacle to investigating ...

Abstract Parkinson’s disease (PD) is a common neurodegenerative disease. Delayed administration of PD medications is associated with increased risk of life-threatening complications including choking, aspiration pneumonia and neuroleptic malignant syndrome. In 2016, the spouse of a patient with PD wrote to Leeds Teaching Hospitals Trust (LTHT) to highlight that multiple medication delays and omissions had occurred during his recent admission. In response, LTHT formed a PD quality improvement (QI) Collaborative ...

Native Americans domesticated maize ( Zea mays ssp. mays ) from lowland teosinte parviglumis ( Zea mays ssp. parviglumis) in the warm Mexican southwest and brought it to the highlands of Mexico and South America where it was exposed to lower temperatures that imposed strong selection on flowering time. Phospholipids are important metabolites in plant responses to low-temperature and phosphorus availability and have been suggested to influence flowering time. Here, we combined linkage mapping with genome ...

Experiments show that elevated atmospheric CO2 (eCO2) often enhances plant photosynthesis and productivity, yet this effect varies substantially and may be climate sensitive. Understanding if, where and how water supply regulates CO2 enhancement is critical for projecting terrestrial responses to increasing atmospheric CO2 and climate change. Here, using data from 14 long-term ecosystem-scale CO2 experiments, we show that the eCO2 enhancement of annual aboveground net primary productivity is sensitive ...

Hypoplastic left heart is a severe human congenital heart defect characterized by left ventricular hypoplasiawith aortic and mitral valve atresia. A genetic etiology is indicated by an association of the hypoplastic left heart phenotype with terminal 11q deletions that span approximately 20 Mb (distal to FRA11B in 11q23). Here we define the breakpoints in four patients with heart defects in association with distal 11q monosomy and refine the critical region to an approximately 9-Mb region distal ...

Abstract CRISPR guide RNA libraries have been iteratively improved to provide increasingly efficient reagents, although their large size is a barrier for many applications. We design an optimised minimal genome-wide human CRISPR-Cas9 library (MinLibCas9) by mining existing large-scale gene loss-of-function datasets, resulting in a greater than 42% reduction in size compared to other CRISPR-Cas9 libraries while preserving assay sensitivity and specificity. MinLibCas9 provides backward compatibility ...

Abstract Determining the extent of dispersal in exploited marine fishes is essential for understanding their population dynamics and optimising management. The West Australian dhufish, Glaucosoma hebraicum , is a highly sought‐after, large and long‐lived reef‐dwelling species, endemic to south‐western Australia. Stock assessments indicate that this indicator species is overexploited. The fishery is managed using a zone‐based system, which implicitly assumes a high degree of demographic independence ...

Background The overall incidence of colorectal cancer is decreasing in many high-income countries, yet analyses in the USA and other high-income countries such as Australia, Canada, and Norway have suggested increasing incidences among adults younger than 50 years. We aimed to examine longitudinal and generational changes in the incidence of colon and rectal cancer in seven high-income countries. Methods We obtained data for the incidence of colon and rectal cancer from 20 population-based cancer ...

Abstract Titanium alloys have a high strength-to-weight ratio, fatigue performance and excellent corrosion resistance, and therefore are widely used in the aerospace sector due to their ability to withstand severe mechanical and thermal stresses in service. There are numerous cases where it would be desirable to use different titanium alloys in defined subcomponent regions to improve performance and efficiency. Conventional processing routes do not permit components to be produced with multiple ...

Background: A new device, Intrabeam, is available for intraoperative radiotherapy. We have prospectively examined its feasibility and tolerability in delivering adjuvant breast cancer treatment. Methods: Thirty‐five patients undergoing breast‐conserving surgery received targeted tumour bed irradiation consisting of 5 Gy (at 10 mm) in a single fraction. This single intraoperative treatment was used to replace the external beam radiotherapy ‘boost’ that would usually be given in 10 daily treatments ...

Rapid morphological change has been shown in rodent populations on islands, including endemic deer mice (Peromyscus maniculatus subspp.) on the California Channel Islands. Surprisingly, most of these changes were towards a smaller size. Black rats were introduced to Anacapa Island in the mid-1800s (probably in 1853) and eradicated in 2001-2002. To assess possible changes in these rats since their introduction, eleven cranial and four standard external measurements were taken from 59 Rattus rattus ...

Rift fault networks can be complex, particularly those developed by multiple periods of non-coaxial extension, comprising non-colinear faults with many interactions. Thus, topology, rather than simple geometry, is required to characterise such networks, as it provides a way to describe the arrangement of individual faults in the network. Topology is analysed here in terms of nodes (isolated I nodes or connected Y or X nodes) and branches (I–I, I–C, C–C branches). In map view, the relative proportions ...
