B-cell chronic lymphocytic leukaemia (B-CLL) is a heterogeneous disease with a highly variable clinical course (Chiorazzi et al, 2005) and several factors at diagnosis predict prognosis and help guide treatment decisions (Seiler et al, 2006). Unmutated immunoglobulin heavy variable (IGHV) genes and high expression of zeta-chain-associated protein kinase 70 (ZAP-70) are well-known factors for poor prognosis (Rassenti et al, 2004). When they are discordant other cytogenetic and molecular biological factors ...

T-cell homeostasis is regulated by several molecules; among these, interleukin (IL)-7 plays an essential role in the survival and homeostatic proliferation of peripheral naive T cells. In a previous study, we investigated whether human mesenchymal stromal cells (MSCs) could be engineered with the IL-7 gene to produce functional level of this cytokine. In the present study, we analyzed the impact of different quantities of IL-7 produced by MSCs on the survival and proliferation of a negative immunoselected ...

Abstract We previously demonstrated that when human bone marrow-derived mesenchymal cells (hMSCs) were co-cultured with CD4+/CD25+ T regulatory cells (Tregs) they maintained the T regulatory phenotype and function over time. Here we studied the TGF-beta/Smad signalling and mitogen-activated protein kinase (MAPK) pathways after Tregs/hMSCs co-culture. After 7 days co-culture of highly purified, immuno-selected Tregs and hMSCs from healthy donors, TGF-beta and IL-10 concentrations were dosed and mRNA ...

Investment in severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) sequencing in Africa over the past year has led to a major increase in the number of sequences that have been generated and used to track the pandemic on the continent, a number that now exceeds 100,000 genomes. Our results show an increase in the number of African countries that are able to sequence domestically and highlight that local sequencing enables faster turnaround times and more-regular routine surveillance. Despite ...

Abstract Despite much investigation into T regulatory cells (Tregs), little is known about the mechanism controlling their recruitment and function. Since mesenchymal cells (MSCs) exert an immune regulatory function and suppress T cell proliferation, this in vitro study investigated their role in Treg recruitment and function. hMSCs and different T cell populations (CD3+, CD3+/CD45RA+, CD3+/CD45RO+, CD4+/CD25+, CD4+/CD25+/CD45RO+, CD4+/CD25+/CD45RA+) from healthy donors were co-cultured for up to 15 ...

Key players in human T-cell development after T- cell-depleted allogeneic stem cell transplants are bone marrow stroma and interleukin-7 (IL-7). We engineered human stromal cells with the IL-7 gene and studied the effects on T cells in vitro and on in vivo immunological reconstitution in NOD/SCID mice. Transduced mesenchymal cells were negative for CD45 and CD14, positive for CD90 (98.15%), CD105 (87.6%), and STRO-1 (86.7%) and stably produced IL-7 (16.37±2SD pg/ml). In cocultures with T cells, IL-7 ...
