Until now, literature data support the fact that the CHOP regimen represents the standard first line treatment for patients with advanced intermediate-grade non-Hcdgkin's lymphoma. Recently, idarubicin has been introduced in clinical trials because of its favourable preclinical profile: it is more active than daunorubicin and doxorubicin against a number of experimental tumour systems and is significantly less cardiotoxic in animal models. From March 1991 to June 1993, 115 previously untreated patients ...

A study is presented on the expression of mitochondrial oxidative phosphorylation complexes in exponentially growing and serum-starved, quiescent human fibroblast cultures. The functional levels of respiratory complexes I and III and complex V (adenosine triphosphate (ATP) synthase) were found to be severely depressed in serum-starved fibroblasts. The depression of oxidative phosphorylation system (OXPHOS) complexes was associated with reduced levels of peroxisome proliferator-activated receptor gamma ...

A study is presented of the kinetics and stoichiometry of fast proton translocation associated to aerobic oxidation of components of the mitochondrial respiratory chain. 1. Aerobic oxidation of ubiquinol and b cytochromes is accompanied in EDTA particles, obtained by sonication of beef-heart mitochondria, by synchronous proton uptake. 2. The rapid proton uptake associated to oxidation and b cytochromes is greatly stimulated by valinomycin plus K+, but is unaffected by carbonyl cyanide p-trifluoromethoxyphenylhydrazone. ...

1. The →H+/e− quotients for proton release from mitochondria associated with electron flow from succinate and duroquinol to O2, ferricyanide or ferricytochrome c, and from NNN′N′-tetramethyl-p-phenylenediamine+ascorbate to O2, were determined from rate measurements of electron flow and proton translocation. 2. Care was taken to avoid, or to take into account, unrelated electron flow and proton translocation, which might take place in addition to the oxido-reductions that were the subject of our analysis. ...

The osmotic properties of EDTA submitochondrial particles have been studied by means of light-scattering measurements and radioisotopic determination of water distribution. It is shown that EDTA particles exhibit a respiration-linked swelling which: (i) requires oligomycin and NO3−; (ii) is promoted by nigericin and inhibited by valinomycin in the presence of K+ but not in the presence of Na+; and (iii) is reversed by FCCP. It is concluded that the energy-linked swelling of EDTA particles is caused ...

The kinetics and mechanism of passive and active proton translocation in submitochondrial vesicles, obtained by sonication of beef heart mitochondria, have been studied. Analysis of the anaerobic release of the protons taken up by submitochondrial particles in the respiring steady state shows that proton diffusion consists of two parallel, apparent first-order processes: a fast reaction which, on the basis of its kinetic properties and response to cations and various effectors, is considered to ...

A study is presented of the effect of chemical modification of lysine amino groups on the redox and protonmotive activity of bovine heart cytochrome c oxidase. Treatment of soluble oxidase with succinic acid anhydride resulted in succinylation of lysines in all the subunits of the enzyme. The consequent change of surface charges from positive to negative resulted in inversion of the orientation of the reconstituted enzyme from right-side-out to inside-out. Reconstitution of the oxidase in phospholipid ...

This paper covers genetic and biochemical aspects of mitochondrial bioenergetics dysfunction in hereditary neurological disorders associated with complex I defects. Three types of hereditary complex I dysfunction are dealt with: (i) homozygous mutations in the nuclear genes NDUFS1 and NDUFS4 of complex I, associated with mitochondrial encephalopathy; (ii) a recessive hereditary epileptic neurological disorder associated with enhanced proteolytic degradation of complex I; (iii) homoplasmic mutations ...

pH changes associated with the mitochondrial cytochrome oxidase reaction with H 2 O 2 have been studied. In the presence of forricyanide or Tris‐phenanthroline complex of Co III as electron acceptors, reaction or H 2 O 2 with the oxidized cytochrome oxidase is accompanied by a steady proton release with a rate constant of ca. 3 M −1 s −1 at pH 6.8. The acidification is completely inhibited by superoxide dismutase and its pro‐steady‐state kinetics correlates with that of the oxoferryl compound (F) accumulation. ...

Presented is a study of the impact on the structure and function of human complex I of three different homozygous mutations in the NDUFS4 gene coding for the 18-kDa subunit of respiratory complex I, inherited by autosomal recessive mode in three children affected by a fatal neurological Leigh-like syndrome. The mutations consisted, respectively, of a AAGTC duplication at position 466–470 of the coding sequence, a single base deletion at position 289/290, and a G44A nonsense mutation in the first ...

The role of supernumerary subunits of bovine heart cytochrome c oxidase has been investigated by examining the effect on the enzymatic activity of limited proteolysis by chymotrypsin, thermolysin, and trypsin. All three proteases, when added to the soluble oxidase, digested subunits III, VIa, and VIb and caused inhibition of electron flow in the oxidase. In addition, trypsin and thermolysin also digested subunit IV. Trypsin cleaved off an N-terminal segment of seven residues; thermolysin cleaved ...

In cytochrome c oxidase, oxido-reductions of heme a/Cu(A) and heme a3/Cu(B) are cooperatively linked to proton transfer at acid/base groups in the enzyme. H+/e- cooperative linkage at Fe(a3)/Cu(B) is envisaged to be involved in proton pump mechanisms confined to the binuclear center. Models have also been proposed which involve a role in proton pumping of cooperative H+/e- linkage at heme a (and Cu(A)). Observations will be presented on: (i) proton consumption in the reduction of molecular oxygen ...

Characterization of two mitochondrial proteins of M 1 42 and 18 kDa, respectively, phosphorylated by the cAMP–dependent protein kinase of bovine heart mitochondria (mtPKA), is presented. A 42 kDa protein is found to be loosely associated to complexes I, III and IV of the respiratory chain and complex V (ATP synthase) in the inner mitochondrial membrane. An 18 kDa protein is associated to complex I in the inner membrane and in a purified preparation of this complex where it can be phosphorylated by ...

ABSTRACT The respiratory chain of mitochondria and bacteria is made up of a set of membrane‐associated enzyme complexes which catalyse sequential, stepwise transfer of reducing equivalents from substrates to oxygen and convert redox energy into a transmembrane protonmotive force ( PMF ) by proton translocation from a negative ( N ) to a positive ( P ) aqueous phase separated by the coupling membrane. There are three basic mechanisms by which a membrane‐associated redox enzyme can generate a PMF . These are membrane ...

The influence of aging on mitochondrial energy metabolism of rat liver and rat heart has been studied by analysis of (i) respiratory rate of succinate-supplemented mitochondria in state III (coupled state in the presence of ADP + Pi); (ii) the rate of synthesis of ATP in succinate-supplemented mitochondria; (iii) the ATP hydrolase activity of sonicated submitochondrial particles. The results indicate a decrease of the F0F1-ATP synthase activity in mitochondria isolated from both organs of aged (24-month- ...

ABSTRACT Proton pumping heme‐copper oxidases represent the terminal, energy‐transfer enzymes of respiratory chains in prokaryotes and eukaryotes. The Cu B ‐heme a 3 (or heme o) binuclear center, associated with the largest subunit I of cytochrome c and quinol oxidases, is directly involved in the coupling between dioxygen reduction and proton pumping. The role of the other subunits is less clear. The following aspects will be covered in this paper:i) the efficiency of coupling in the mitochondrial ...
