Integrins are a family of heterodimeric receptors that bind to components of the extracellular matrix and influence cellular processes as varied as proliferation and migration. These effects are achieved by tight spatiotemporal control over intracellular signalling pathways, including those that mediate cytoskeletal reorganisation. The ability of integrins to bind to ligands is governed by integrin conformation, or activity, and this is widely acknowledged to be an important route to the regulation ...

We report the mode-locked operation of two new Yb-doped oxyorthosilicates, Y2SiO5 (YSO) and Lu2SiO5 (LSO), that are longitudinally diode pumped. Yb:YSO supplied pulses as short as 122 fs with 410 mW of output power at 1041 nm. More than 2.6 W of average output power, for pulse durations of 198 fs at 1044 nm and 260 fs at 1059 nm for Yb:YSO and Yb:LSO, respectively, were provided. These are, to our best knowledge, the highest values ever obtained and the most efficient mode-locked laser in such a classic ...

This review discusses recent advances in our understanding of adhesion receptor trafficking in vitro, and extrapolates them as far as what is currently possible towards an understanding of migration in three dimensions in vivo. Our specific focus is the mechanisms for endocytosis and recycling of the two major classes of cell-matrix adhesion receptors, integrins and syndecans. We review the signalling networks that are employed to regulate trafficking and conversely the effects of trafficking on ...

We report what is believed to be the first demonstration of a high-power passively mode-locked diode-pumped femtosecond laser based on an Yb3+:CaF2 single crystal, directly pumped by a 15-W fiber-coupled laser diode. With a 5-at. % Yb3+ -doped sample and prisms for dispersion compensation we obtained pulses as short as 150 fs, with 880 mW of average power and up to 1.4-W average output power, with a pulse duration of 220 fs, centered at 1049 nm. The laser wavelength could be tuned from 1040 to 1053 ...

Results of diode-pumped cw laser operation of an Yb3+:CaF2 single crystal are reported for what is to our knowledge the first time. With a 5-at. % Yb3+-doped sample we obtained 5.8-W output power at 1053 nm for 15 W of incident power at 980 nm. The laser wavelength could be tuned from 1018 to 1072 nm, and a small-signal gain as high as 1.8 was achieved, showing the great potential of Yb3+:CaF2 as an amplifier medium for femtosecond pulses.

Atrial fibrillation (AF) is the most common form of cardiac arrhythmia. Surgical/Radiofrequency (RF) ablation is a therapeutic procedure that consists of creating lines of conduction block to interrupt AF. The present study evaluated 13 different ablation patterns by means of a biophysical model of the human atria. In this model, ablation lines were abruptly applied transmurally during simulated sustained AF, and success rate, time to AF termination and average beat-to-beat interval were documented. ...

This work reports single-frequency laser oscillation at λ=1003.4 nm of a diode-pumped vertical external cavity surface-emitting semiconductor laser for metrological applications. A low thermal resistance of the semiconductor active component is achieved by solid-liquid interdiffusion bonding onto a SiC substrate. The spectro-temporal dynamics of the laser is theoretically studied. Experimentally, an output power of 1.7 W is demonstrated in free running operation, and up to 500 mW in a true single ...

Single-frequency lasers in the blue-green (500 nm) visible spectrum are important for many applications including iodine precision spectroscopy, optical frequency standards or ionized argon lasers substitution. To the best of our knowledge, we report in this paper the first single-frequency diode-pumped Ytterbium-doped solid-state laser operating at 501.7 nm by intracavity second harmonic generation (SHG). The single-frequency fundamental infrared laser light is generated by a diode-pumped Yb3+:Y2SiO5 ...

We report here on the efficient laser action of a new Yb-doped crystal Yb3+:Lu2SiO5 (Yb:LSO) under high power diode-pumping (15 W). Its performances were compared to another Yb-doped crystal belonging to the oxyorthosilicate family: the Yb:YSO. For both crystals, more than 7 W of laser radiation around 1 μm was obtained under 14.4 W of incident pump power at 978 nm, leading to high optical conversions of more than 50%. Finally, both crystals demonstrate little sensitivity to pump wavelength drift ...

Cell migration makes a fundamental contribution to both normal physiology and disease pathogenesis. Integrin engagement with extracellular ligands spatially controls, via the cyclical activation and deactivation of the small GTPase Rac1, the dynamic membrane protrusion and cytoskeletal reorganization events that are required for directional migration. While the pathways that control integrin-mediated Rac1 activation are reasonably well defined, the mechanisms that are responsible for switching off ...

Inhibition of αvβ3 or expression of mutant p53 promotes invasion into fibronectin (FN)-containing extracellular matrix (ECM) by enhancing Rab-coupling protein (RCP)–dependent recycling of α5β1 integrin. RCP and α5β1 cooperatively recruit receptor tyrosine kinases, including EGFR1, to regulate their trafficking and downstream signaling via protein kinase B (PKB)/Akt, which, in turn, promotes invasive migration. In this paper, we identify a novel PKB/Akt substrate, RacGAP1, which is phosphorylated as ...

Invasive migration in 3D extracellular matrix (ECM) is crucial to cancer metastasis, yet little is known of the molecular mechanisms that drive reorganization of the cytoskeleton as cancer cells disseminate in vivo. 2D Rac-driven lamellipodial migration is well understood, but how these features apply to 3D migration is not clear. We find that lamellipodia-like protrusions and retrograde actin flow are indeed observed in cells moving in 3D ECM. However, Rab-coupling protein (RCP)-driven endocytic ...

This work reports single-frequency laser oscillation at 1003.4 nm of an optically pumped external cavity semiconductor laser. By using a gain structure bonded onto a high conductivity substrate, we demonstrate both theoretically and experimentally the strong reduction of the thermal resistance of the active semiconductor medium, resulting in a high power laser emission. The spectro-temporal dynamics of the laser is also explained. Furthermore, an intracavity frequency-doubling crystal was used to ...

Abstract Bacterial two-component systems (TCSs) are signaling modules that control physiology, adaptation, and host interactions. A typical TCS consists of a histidine kinase (HK) that activates a response regulator via phosphorylation in response to environmental signals. Here, we systematically test the effect of inactivating the conserved phosphatase activity of HKs to activate TCS signaling pathways. Transcriptome analyses of 14 HK mutants in Streptococcus agalactiae , the leading cause of neonatal ...

Abstract Streptococcus agalactiae is among the few pathogens that have not developed resistance to ß-lactam antibiotics despite decades of clinical use. The molecular basis of this long-lasting susceptibility has not been investigated, and it is not known whether specific mechanisms constrain the emergence of resistance. In this study, we first report ß-lactam tolerance due to the inactivation of the c-di-AMP phosphodiesterase GdpP. Mechanistically, tolerance depends on antagonistic regulation by ...

This crystal exhibits good thermo-optical properties and can easily be grown in bulk crystals or in thin films. A maximum power of 5.8 W in a diffraction limited beam has been obtained with a 5% ytterbium-doped crystal of 4 mm-long. Moreover, the laser wavelength has been tuned over 54 nm, between 1018 and 1072 nm, and the double-pass small-signal gain has been measured to be more than ...
