After the war Piotr wanted to pursue a career in research and moved to France to work with Boris Ephrussi. Ephrussi was not always an easy person to work with, but Piotr worked closely with him for many years, which earned him the title of 'Le mutant résistant' from the molecular biologists of the Institute Pasteur. After a brief and unsuccessful period working on development in sea urchin eggs, Piotr began his work with yeast, studying 'petite' mutants. This was the beginning of Piotr's studies on ...

Memoirs are more often written by generals than by scientists. Although the highest rank I received during my five years in the Polish Underground Army, Armia Krajowa, did not exceed that of a corporal-chief (not a very brilliant accomplishment!), it would have been much easier, and possibly even more interesting for the readers, to write about this period. At least there was action, blood, fantastic friends and beautiful girls: a more-or-less classical potpourri of cliches that a survivor (who, in ...

The kinetics of inactivation by Versene of d-lactate dehydrogenase from anaerobic yeast was studied. Inactivation is irreversible: a diminution of free Versene concentration down to 10−14M does not lead to any reactivation. The initial velocity of inactivation is proportional to Versene concentrations and to enzyme concentrations. The Michaelis constant (in the reaction d-lactate to pyruvate) of the partially inactivated enzyme is not modified. Activation energy of inactivation is low (⩽ 3kcal/mole). ...

A method is described for a quantitative extraction, separation and determination of two distinct molecular species of cytochrome c from yeast: iso-1-cytochrome c and iso-2-cytochrome c. This method is applicable, with an overall yield of about 90 %, to small quantities of yeast (about 2 g) and small quantities of cytochrome c (about 5 mμmoles). The time course of biosyntheses of two isocytochromes c was simultaneously studied during induction by O2 and found different. Immediately upon admission of ...

The usefulness of mutants in the unraveling of complex, highly organized, membrane-bound processes such as oxidative phosphorylation is illustrated by a study of a single recessive gene mutation in yeast, designated op(1), which has abolished the efficiency in vivo and in vitro of oxidative phosphorylation without impairing the electron transfer.

Abstract A thorough formal investigation of pair correlations between amino acids in various protein sequences is performed. To this end we apply a particularly sensitive method of statistical analysis. An evidence is given for the existence of correlations between amino acids separated, along the polypeptide chain, by 5 and 6 and by 28 and 29 peptide bonds. We discuss the chemical nature of these correlations and their possible relation to the three-dimensional structure of proteins.

Spatial resolution of environmental data may influence the results of habitat selection models. As high-resolution data are usually expensive, an assessment of their contribution to the reliability of habitat models is of interest for both researchers and managers. We evaluated how vegetation cover datasets of different spatial resolutions influence the inferences and predictive power of multi-scale habitat selection models for the endangered brown bear populations in the Cantabrian Range (NW Spain). ...

A method for the estimation of cytochrome concentrations in whole yeast cells was worked out with strains of Saccharomyces cerevisiae obtained in the laboratory. Differences of cytochrome c content were due to mutations affecting the nature or the quantity of synthetized cytochrome c . Respiratory deficient mutants (without cytochromes a + a 3 and b ) were also used. Spectra were measured directly on yeast pastes (up to 3 mm thick) with a Cary 15 recording spectrophotometer provided with a Xenon lamp. Absorption ...

The mitochondrial DNA's from two “grande” yeast strains and three cytoplasmic “petite” mutants were isolated by hydroxyapatite chromatography and investigated in their chemical and physical properties. The buoyant densities of all DNA's were equal to 1.683 g/cm3, except for one from a petite strain which had a density equal to 1.678 g/cm3. The mitochondrial DNA's from the petite mutants were “reversible”, their buoyant densities and their ultraviolet absorptions showing very little or no increase after ...

Yeast is a facultative anaerobe and uses diverse electron acceptors to maintain redox-regulated import of cysteine-rich precursors via the mitochondrial intermembrane space assembly (MIA) pathway. With the growing diversity of substrates utilizing the MIA pathway, understanding the capacity of the intermembrane space (IMS) to handle different types of stress is crucial. We used MS to identify additional proteins that interacted with the sulfhydryl oxidase Erv1 of the MIA pathway. Altered inheritance ...

Ion‐exchange chromatography of the total cytochrome c extracted from a wild‐type laboratory strain of Saccharomyces cerevisiae shows the presence of two minor cytochrome c peaks, besides the already known iso‐1 and iso‐2 cytochromes c. Their amino acid composition is very similar to that of iso‐1, but one of them lacks the ɛ‐ N ‐trimethylated lysine residue characteristic of yeast cytochromes c , thus excluding, at least for this peak, an artifactual origin.
