S ummary One‐hour incubation of reticulocytes with 10 −2 m isonicotinic acid hydrazide (INH) and transferrin‐bound 59 Fe changes the normal distribution of radioiron inside the cell. About 10% of 59 Fe is found in haem and 90% is present in the non‐haem iron pool. The accumulated non‐haem radioiron may be utilized for haem synthesis. Abstract In the presence of hemin 59Fe incorporation into both heme and reticulocytes decreases. Isonicotinic acid hydrazide which inhibits heme synthesis in vitro is shown to increase reticulocyte radioiron uptake. During inhibition of globin synthesis by cycloheximide, reticulocyte radioiron uptake decreases. A method of lead analysis by the cathodic stripping voltammetry technique is suggested in the present study. The experimental conditions and the influence of these factors on the behaviour of lead and sensitivity of the analysis are discussed. This paper reviews and reports the results of experiments on the mechanism by which iron is delivered from extracellular transferrin to reticulocyte mitochondria in which haem is synthesized. Filling of cavities in the uterine wall during hysterography was observed in 54 of 320 surgically excised specimens in which metal threads had been inserted at different levels for identification. Iron compartments in which iron accumulates during the inhibited heme synthesis after treatment with isonicotinic acid hydrazide were studied in rabbit reticulocytes. A great accumulation of 59Fe radioactivity was found in mitochondria, low molecular weight iron compounds and non-hemoglobin proteins, especially ferritin. Incubation of rabbit reticulocytes with cycloheximide and 59Fe bound to transferrin in plasma induces excessive non-hemoglobin 59Fe-labeled heme accumulation in mitochondria. During incubation of these mitochondria in vitro a part of 59Fe-labeled heme is released into the surrounding medium. The addition of globin or bovine serum albumin to the incubation mixture essentially increases the amount of heme released from mitochondria.

S ummary . When isoniazid and pyridoxal are mixed in equimolar quantities a hydrazone is formed which is able to complex with iron. The oral administration of this compound to rats in single doses of 25–100 mg/kg leads to an increase in faecal iron excretion up to 8 times the normal level. In tissue culture the compound is able to remove iron from Chang cells. The results suggest that this compound may be of potential value for the oral therapy of iron overload.

We have investigated the effect of succinylacetone (4,6-dioxoheptanoic acid) on hemoglobin synthesis and iron metabolism in reticulocytes.
