S ummary One‐hour incubation of reticulocytes with 10 −2 m isonicotinic acid hydrazide (INH) and transferrin‐bound 59 Fe changes the normal distribution of radioiron inside the cell. About 10% of 59 Fe is found in haem and 90% is present in the non‐haem iron pool. The accumulated non‐haem radioiron may be utilized for haem synthesis. This is demonstrated by the reincubation of washed reticulocytes with a high non‐haem radioiron pool induced by INH under optimal conditions. The incorporation of radioiron ...

Abstract In the presence of hemin 59Fe incorporation into both heme and reticulocytes decreases. Isonicotinic acid hydrazide which inhibits heme synthesis in vitro is shown to increase reticulocyte radioiron uptake. During inhibition of globin synthesis by cycloheximide, reticulocyte radioiron uptake decreases. This decrease is probably caused by free heme which accumulates in reticulocytes after inhibition of globin synthesis. Simultaneous addition of an inhibitor of heme synthesis—isonicotinic ...

A method of lead analysis by the cathodic stripping voltammetry technique is suggested in the present study. The experimental conditions and the influence of these factors on the behaviour of lead and sensitivity of the analysis are discussed. The use of the rotating platinum electrode and of a derivative circuit is found to be useful. Compared with amalgam polarography, the cathodic stripping technique has the advantage of easy separation from copper, cadmium, zinc, etc., although the amalgam polarography ...

The problem of the feedback inhibitory effect of heme on its own synthesis in erythroid cells was studied using different methods. The activity of δ‐aminolevulinic acid synthetase was determined in the insoluble particles containing mitochondria and stroma prepared from rabbit reticulocytes and in the lysate from these reticulocytes. The effect of hemin on the incorporation of [2‐ 14 C]glycine into heme was measured in the lysate from rabbit reticulocytes. The effect of hemin on the incorporation ...

Abstract 1. 1. All of the tested inhibitors of globin synthesis (puromycin, cycloheximide, emetine, chloramphenicol, tryptamine, NaF, amino acid deficit) decrease reticulocyte radio-iron uptake. Experimental evidence showed that the mechanism of inhibited incorporation of iron into reticulocytes differed according to the inhibitor used. 2. 2. The direct inhibitors of globin synthesis inhibit the membrane transport of iron by a mechanism which depends on the ability of cells to form heme. This is ...

In the hemolysate of reticulocytes incubated with 59Fe or [2–14C]glycine a radioactivity is found also in nonhemoglobin heme. About 96% of the radioactivity of nonhemoglobin heme is associated with proteins and approx. 4% is in a free form. Labeled free heme is removed from the reticulocyte hemolysate on the column of Sephadex G-25. Retained heme is eluted from the column of Sephadex G-25 by means of the special eluting solution containing serum albumin. Protein-bound nonhemoglobin heme radioactivity ...

This paper reviews and reports the results of experiments on the mechanism by which iron is delivered from extracellular transferrin to reticulocyte mitochondria in which haem is synthesized. It is suggested that transferrin donates the iron directly to mitochondria. Transferrin seems to be bound to mitochondria during the process of iron release. When the release of iron from transferrin is blocked by haem, the iron-transferrin complex remains bound to mitochondria so that the total amount of transferrin ...

Filling of cavities in the uterine wall during hysterography was observed in 54 of 320 surgically excised specimens in which metal threads had been inserted at different levels for identification. Adenomyosis may have accounted for these cavities in 24%. Cavities secondary to adenomyosis in one part of the uterus coexisted when cavities not etiologically related to the disease in another part. Adenomyosis is generally not involved in the pathogenesis of cavities in the lower half of the uterus and ...

S ummary Biliary excretion of iron after administration of pyridoxal isonicotinoyl hydrazone (PIH), a recently identified effective iron‐chelating agent, was investigated in rats. PIH administered both intraperitoneally and orally was shown to increase significantly 59 Fe excretion into bile of rats which had previously been injected with 59 Fe‐transferrin to label hepatic parenchymal cells. 59 Fe‐PIH appears in bile as early as 15 min after chelator administration and the peak of 59 Fe‐radioactivity ...

Iron compartments in which iron accumulates during the inhibited heme synthesis after treatment with isonicotinic acid hydrazide were studied in rabbit reticulocytes. A great accumulation of 59Fe radioactivity was found in mitochondria, low molecular weight iron compounds and non-hemoglobin proteins, especially ferritin. In a chase experiment, approximately 50% of the 59Fe radioactivity accumulated in mitochondria and low molecular weight iron compounds was re-utilized for the synthesis of hemoglobin. ...

Incubation of rabbit reticulocytes with cycloheximide and 59Fe bound to transferrin in plasma induces excessive non-hemoglobin 59Fe-labeled heme accumulation in mitochondria. During incubation of these mitochondria in vitro a part of 59Fe-labeled heme is released into the surrounding medium. The addition of globin or bovine serum albumin to the incubation mixture essentially increases the amount of heme released from mitochondria.

δ-Amino [4-14C]laevulinate added to reticulocytes incubated in vitro is incorporated into haem. Exogenous δ-aminolaevulinate restores the incorporation of 59Fe into haem in reticulocytes which had been treated with isonicotinic acid hydrazide (INH) or penicillamine and were hence unable to synthesize δ-aminolaevulinate. On the other hand, the addition of δ-aminolaevulinate does not restore the incorporation of Fe into reticulocytes incubated with haemin. The inhibition of the incorporation of iron ...

The concentration of radioactivity in the thyroid was higher than the average concentration as early as 5 min. after injection and it remained higher than in any other organ during the 4 days observation period. A high radioactivity was also found in the walls of large blood vessels, adrenal ...

S ummary . When isoniazid and pyridoxal are mixed in equimolar quantities a hydrazone is formed which is able to complex with iron. The oral administration of this compound to rats in single doses of 25–100 mg/kg leads to an increase in faecal iron excretion up to 8 times the normal level. In tissue culture the compound is able to remove iron from Chang cells. The results suggest that this compound may be of potential value for the oral therapy of iron overload.

Rabbit reticulocytes with a high level of non-heme radioiron induced by preincubation with isonicotinic acid hydrazide and transferrin-bound 59Fe, were reincubated with various synthetic chelating agents and the amount of radioiron released from the cells was determined. Some substances, especially derivatives of pyridoxal or 2-hydroxybenzaldehyde and isonicotinic acid hydrazide or benzhydrazide, were found to mobilize significantly iron from 59Fe-labeled reticulocytes. We have investigated the effect of succinylacetone (4,6-dioxoheptanoic acid) on hemoglobin synthesis and iron metabolism in reticulocytes. Succinylacetone, 0.1 and 1 mM, inhibited [2-14C]glycine incorporation into heme by 91.2 and 96.4%, respectively, and into globin by 85 and 90.2%, respectively. 60 μM hemin completely prevented the inhibition of globin synthesis by succinylacetone, indicating that succinylacetone inhibits specifically the synthesis of heme. Added porphobilinogen, but not δ-aminolevulinic ...
