Abstract Purpose To implement and evaluate a novel single‐volume two‐dimensional localized constant‐time‐based correlated spectroscopy (2D LCT‐COSY) sequence on a clinical 3T MR scanner. This sequence exhibits homonuclear decoupling along the F1 dimension, leading to improved spectral resolution compared to that of non‐constant‐time localized correlated spectroscopy (L–COSY). Materials and Methods A GE 3T MR scanner equipped with a quadrature transmit and receive extremity coil was used in this study. ...

Homing endonucleases are highly specific catalysts of DNA strand breaks that induce the transposition of mobile intervening sequences containing the endonuclease open reading frame. These enzymes recognize long DNA targets while tolerating individual sequence polymorphisms within those sites. Sequences of the homing endonucleases themselves diversify to a great extent after founding intron invasion events, generating highly divergent enzymes that recognize similar target sequences. Here, we visualize ...

Abstract Assessments of safety and efficacy are crucial before human ESC (hESC) therapies can move into the clinic. Two important early potential hESC applications are the use of retinal pigment epithelium (RPE) for the treatment of age-related macular degeneration and Stargardt disease, an untreatable form of macular dystrophy that leads to early-onset blindness. Here we show long-term functional rescue using hESC-derived RPE in both the RCS rat and Elov14 mouse, which are animal models of retinal ...

The LAGLIDADG homing endonucleases include free-standing homodimers, pseudosymmetric monomers, and related enzyme domains embedded within inteins. DNA-bound structures of homodimeric I-CreI and monomeric I-SceI indicate that three catalytic divalent metal ions are distributed across a pair of overlapping active sites, with one shared metal participating in both strand cleavage reactions. These structures differ in the precise position and binding interactions of the metals. We have studied the metal ...

The crystal structure of the human B blood group related trisaccharide α‐ l ‐Fuc p ‐(1→2)‐[α‐ d ‐Gal p ]‐(1→3)‐β‐ d ‐Gal p ‐OCH 3 ( 1 ) has been determined. The solution structure of 1 was studied by two‐dimensional NMR techniques at 600 MHz in D 2 O solution and the conformational properties were analyzed in terms of the torsional angles φ H and ψ H , derived from 3 J CH coupling constants, and 10 inter‐ residue proton–proton distances. 3 J CH could be accurately measured by a recently introduced two‐dimensional ...

A recently developed technique for the probing of the combining sites of lectins and antibodies, to establish the structure of the epitope that is involved in the binding of an oligosaccharide, is used to study the binding of methyl α-isomaltoside by the enzyme glucoamylase. The procedure involved the determination of the effects on the kinetics of hydrolysis of both monodeoxygenation and mono-O-methylation at each of the seven hydroxyl groups in order to gain an estimate of the differential changes ...
