The importance of the amyloid precursor protein (APP) in the pathogenesis of Alzheimer’s disease (AD) became apparent through the identification of distinct mutations in the APP gene, causing early onset familial AD with the accumulation of a 4-kDa peptide fragment (βA4) in amyloid plaques and vascular deposits. However, the physiological role of APP is still unclear. In this work, Drosophila melanogaster is used as a model system to analyze the function of APP by expressing wild-type and various mutant ...

PF-06747775 (PF-7775) is a highly potent, selective third generation irreversible EGFR-TKI, effective against EGFR-TKI sensitizing and resistance (T790M) mutations in NSCLC cell lines; IC50s between 3-12 nM and 26X greater selectivity toward mutant vs. wild-type (WT) EGFR. This is the first report from an ongoing phase I, first in human multicenter study (NCT02349633) of PF-7775 in patients with metastatic EGFRm+ NSCLC. EGFRm+ NSCLC pts, with acquired resistance to EGFR-TKIs enrolled into dose escalation ...

Background: PF-06747775 ("PF-7775") is a selective, 3rd generation Epidermal Growth Factor Receptor (EGFR) TKI effective against EGFR sensitizing and T790M mutations in preclinical models. Enrollment was completed in a phase I study of PF-7775 in EGFR mutant (EGFRm+) NSCLC patients (pts) who progressed after a prior EGFR TKI. Methods: EGFRm+ NSCLC pts with resistance to first-line EGFR-TKIs enrolled in a multicenter trial in 6 dose escalation (25 mg–600 mg) and 2 dose expansion cohorts (200 mg and 300 ...

The acceptance of master–slave robotic teleoperated applications in the medical field is related not only to the accuracy and precision of the robotic systems but also to the haptic features. Indeed, the capability to render a good haptic feeling, hence the sensation to drive the real surgical tool, is necessary for reaching an effective interaction between surgeon and robotic system. In this paper an innovative controller for master–slave haptic systems for neurosurgery has been developed by getting ...

Symmetric encryption is the most widely adopted security solution for wireless sensor networks. The main open issue in this context is represented by the establishment of symmetric keys. Although many key management schemes have been proposed in order to guarantee a high security level, a solution without weaknesses does not yet exist. An important class of key management schemes is based on a transitory master key (MK). In this approach, a global secret is used during the initialization phase to ...

Background: Mental fatigue is a psychobiological state caused by extended periods of cognitive effort, and evidence suggests that mentally fatigued athletes present impaired physical performance. Different ergogenic aids have been proposed to counteract the deleterious effects of mental fatigue, but whether brain stimulation can counteract mental fatigue is still unknown. This scenario is even more obscure considering the effects of these interventions (mental fatigue induction and brain stimulation) ...

We defined the role of the transcriptional factor–XYR1–in the filamentous fungus Trichoderma reesei during cellulosic material degradation. In this regard, we performed a global transcriptome analysis using RNA-Seq of the ∆xyr1 mutant strain of T. reesei compared with the parental strain QM9414 grown in the presence of cellulose, sophorose, and glucose as sole carbon sources. We found that 5885 genes were expressed differentially under the three tested carbon sources. Of these, 322 genes were upregulated ...

This paper presents a new force-reflecting control system for master–slave haptic devices. This controller has been implemented and tested on the robotic systems for minimally invasive neurosurgery developed by our Research Group. Robot-assisted surgery is a very valuable treatment, since it allows benefits of high precision, accuracy, and repeatability of robotic devices. The proposed controller is meant to be used for master–slave haptic robotic surgery, but it can be used for any device that ...

This study describes a simple, rapid, and cost-effective fabrication of PDMS electrophoresis microchips using poly(vinyl acetate) (PVAc) emulsion as photoresist master. High-relief microfluidic structures were defined on poly(vinyl acetate) previously deposited on printed circuit boards surfaces without cleanroom facilities and sophisticated instrumentation. After a UV exposure, channels with heights ranging from 30 to 140 μm were obtained by controlling the emulsion mass deposited on the master ...

Purpose: To compare in a real clinical setting the largely unknown midterm clinical effectiveness of two protease inhibitor (PI)-based highly active antiretroviral therapy (HAART) regimens with different potency and tolerability profiles in naïve patients. Method: This study was a multicenter, open-label, randomized trial in naïve patients with less than 400 CD4+ cell count/μL, regardless of viral load. Treatment arms were hard gel capsule saquinavir (HGC-SQV)-based HAART (Arm A), with an expected more ...
