An immunoperoxidase procedure was employed to study the expression of a large-molecular-weight, virus-induced polypeptide (VP175; molecular weight, 175,000) at the light and electron microscopic levels in Vero cells infected with herpes simplex virus type 1 or with tsB2, a DNA-negative, temperature-sensitive mutant of herpes simplex virus type 1. In cells infected with herpes simplex virus type 1 and in cells infected with tsB2 at the permissive temperature (34 degrees C), VP175 was found within the ...

Abstract Osteosarcoma (OS) is an aggressive sarcoma, where novel treatment approaches are required. Genomic studies suggest that a subset of OS, including OS tumour cell lines (TCLs), exhibit genomic loss of heterozygosity (LOH) patterns reminiscent of BRCA1 or BRCA2 mutant tumours. This raises the possibility that PARP inhibitors (PARPi), used to treat BRCA1/2 mutant cancers, could be used to target OS. Using high-throughput drug sensitivity screening we generated chemosensitivity profiles for 79 small ...

We present the results of prompt optical follow-up of the electromagnetic counterpart of the gravitational-wave event GW170817 by the Transient Optical Robotic Observatory of the South Collaboration (TOROS). We detected highly significant dimming in the light curves of the counterpart (Delta g=0.17+-0.03 mag, Delta r=0.14+-0.02 mag, Delta i=0.10 +- 0.03 mag) over the course of only 80 minutes of observations obtained ~35 hr after the trigger with the T80-South telescope. A second epoch of observations, ...

In early 2014, the UK Department for Transport (DfT) commissioned the first national value of travel time (VTT) study since the mid-1990s. This paper presents the methodological work undertaken for this study, with important innovations along a number of dimensions, both in terms of survey design and modelling methodology. Our findings show a rich pattern of heterogeneity across the travelling public, in terms of an impact on the VTT by both person and trip characteristics, as well as a major role ...

This paper identifies, relates and compares two popular modelling approaches to estimate the value of travel time changes. The first (random utility [RU]) assumes that the random component of the model relates to the difference between the utilities of travel options; the second (random valuation [RV]) assumes that it relates to the difference between the value of travel time and a suggested valuation threshold. This paper gives details of the theoretical relationship between the two approaches and ...

Abstract: The characteristic rash of systemic juvenile idiopathic arthritis is a transient erythematous eruption associated with a quotidian spiking fever. Usually asymptomatic, it can be pruritic, with dermatographism at sites of scratching or pressure. An illness similar to this entity in adults is designated adult‐onset Still disease. The relationship between the pediatric and adult disease is uncertain and differences in case definition have evolved. Specifically, a sustained arthritis for at ...

The role of conformation-based quality control in the early secretory pathway is to eliminate misfolded polypeptides and unassembled multimeric protein complexes from the endoplasmic reticulum, ensuring the deployment of only functional molecules to distal sites. The intracellular fate of terminally misfolded human α 1 -antitrypsin was examined in hepatoma cells to identify the functional role of asparagine-linked oligosaccharide modification in the selection of glycoproteins for degradation by the ...

The potassium channel from Streptomyces lividans is an integral membrane protein with sequence similarity to all known K + channels, particularly in the pore region. X-ray analysis with data to 3.2 angstroms reveals that four identical subunits create an inverted teepee, or cone, cradling the selectivity filter of the pore in its outer end. The narrow selectivity filter is only 12 angstroms long, whereas the remainder of the pore is wider and lined with hydrophobic amino acids. A large water-filled ...

Protection of lung elastin fibers from proteolytic destruction is compromised by inefficient secretion of incompletely folded allelic variants of human α1-antitrypsin from hepatocytes. Pulse-chase radiolabeling with [35S]methionine and sucrose gradient sedimentation and coimmunoprecipitation techniques were employed to investigate quality control of human α1-antitrypsin secretion from stably transfected mouse hepatoma cells. A novel means of extracting carboxylic acids from aqueous solutions is described which involves the use of a lipolytic enzyme to convert the acid to a more hydrophobic ester. A water-immiscible long-chain alcohol serves both as a reactant in esterifying the acid, and as a solvent for extraction of the ester. Radiochemical tracer studies of the underlying reaction and extraction equilibria established that a low equilibrium concentration of ester (0.04-0.5mM) in the presence of water is counterbalanced ...

Most bio-economic models in fisheries assume perfectly rational profit-maximizing behaviour by fishing vessels. Here we investigate this assumption empirically. Using a flexible agent-based model of fishing vessels called POSEIDON, we compared predicted fishing patterns to observed patterns in logbook data, that resulted from a wide range of stylized decision-making processes in the U.S. west coast dover sole-thornyhead-sablefish (DTS) fishery, which is managed with tradable quotas (ITQs). The cDNA for the PPi-dependent phosphofructo-1-kinase has been cloned and sequenced from a cDNA library prepared from the free-living amoeba Naegleria fowleri. The coding sequence of the cDNA consists of 1311 bases which translates into 437 amino acids with a molecular mass of 48095 Da. Comparison of the sequence with those of the previously described sequences of PPi-dependent phosphofructokinases from Propionibacterium freudenreichii and potato tuber revealed amino acid identities of 23 and 28% ...

The relief of symptoms of myocardial ischemia and its lasting effect may be related to completeness of myocardial revascularization. Multiple bypass grafting to arteries with lesser stenosis and to small arteries can be simplified by use of the sequential technic. This can be done with no added risk, but its ultimate effectiveness will have to be determined at a much later date.

ZnO is a wide bandgap semiconductor in which point and extended defects tune its optoelectronic properties, and the identification of essential microscopic ingredients for such a tunning is a challenging task. In this work, we combine experimental techniques with theoretical calculations at the atomistic level to investigate a variety of neutral and charged point defects along with their response to thermal treatment by optical and transport measurements. We obtain photoluminescence spectra compatible ...

Parkinson's disease (PD) is a progressive and currently incurable neurological disorder characterised by the loss of midbrain dopaminergic neurons and the accumulation of aggregated alpha-synuclein (a-syn). Oligomeric a-syn is proposed to play a central role in spreading protein aggregation in the brain with associated cellular toxicity contributing to a progressive neurological decline. For this reason, a-syn oligomers have attracted interest as therapeutic targets for neurodegenerative conditions ...

Abstract Environmental remediation efforts in low‐ and middle‐income countries have yet to be evaluated for their cost effectiveness. To address this gap we calculate a cost per Disability Adjusted Life Year (DALY) averted following the environmental remediation of the former lead smelter and adjoining residential areas in Paraiso de Dios, Haina, the Dominican Republic, executed from 2009 to 2010. The remediation had the effect of lowering surface soil lead concentrations to below 100 mg/kg and measured ...

Cognition is a major subject to be addressed nowadays due to the increasing number of cognitively affected people in most societies. Because of a lack of pharmaceutical therapies treating cognitive decline, its indicators should be diagnosed before it becomes prevalent. Scientific evidence indicates a relationship between cognition and the nervous system, especially its autonomic part. Heart rate variability (HRV) as an indicator of the autonomic nervous system functioning has been studied as a biological ...
