The incorporation of [methyl- 3 H]thymidine (TdR) into bacterial DNA in Hartbeespoort Dam, South Africa was measured over 16 mo and at nine depths. Bacterial numbers at the surface ranged between 2.45 and 32.20 × 10 6 cells∙mL −1 [Formula: see text] while bacterial production varied between 1.0 and 251 pmol TdR∙L −1 ∙h −1 (0.01 to 1.9 mg C∙m −3 ∙h −1 ). At the bottom, production ranged between 0 and 26.7 pmol TdR∙L −1 ∙h −1 (0–0.2 mg C∙m −3 ∙h −1 ). The fastest bacterial doubling time was 59 h. At the surface, bacterial ...

The incorporation of [ methyl - 3 H]thymidine into three macromolecular fractions, designated as DNA, RNA, and protein, by bacteria from Hartbeespoort Dam, South Africa, was measured over 1 year by acid-base hydrolysis procedures. Samples were collected at 10 m, which was at least 5 m beneath the euphotic zone. On four occasions, samples were concurrently collected at the surface. Approximately 80% of the label was incorporated into bacterial DNA in surface samples. At 10 m, total incorporation of label ...

It is essential during measurements of aquatic bacterial production with [methyl#x2010; 3 H]thymidine (Tdr) that only labeled DNA is measured. We found in 12 freshwater and marine systems that DNA labeling represented a variable proportion of total macromolecular labeling. Up to 87% of label appearing in precipitated labeled macromolecules from acid#x2010;base hydrolysis treatments was soluble in ethanol. Reverse#x2010;phase, high#x2010;pressure liquid chromatography showed that the composition of ...

We evaluated the potential of prairie wetlands in North America as carbon sinks. Agricultural conversion has resulted in the average loss of 10.1 Mg ha(-1) of soil organic carbon on over 16 million ha of wetlands in this region. Wetland restoration has potential to sequester 378 Tg of organic carbon over a 10-year period. Wetlands can sequester over twice the organic carbon as no-till cropland on only about 17% of the total land area in the region. We estimate that wetland restoration has potential ...

Primary productivity, using the 14C technique was measured in the expanded lake-like upper reaches of a South African estuary during 1972. Maximal assimilation per unit volume ranged between 5.0 and 13.1 mg C m−3 h−1 and areal productivity ranged between 13.1 and 36.9 mg C m−2 h−1. For most of the period the euphotic zone (level of 1% of surface light) extended to only 4.5 m. The poor light conditions in the estuary were due to the heavy content of humic material and suspended detrital matter. Light, ...

SUMMARY A comparison was made between acridine orange (AO) and 4'6-diamidino-2-phenylindole (DAPI), a highly sensitive DNA fluorescing stain, for enumerating aquatic bacteria. When water samples were processed in a darkened room and a final DAPI concentration of 0,05 μg ml −1 was used, there was no significant difference between the population estimates obtained with AO and DAPI. With DAPI, bacteria are more easily distinguished from other particulate matter than when stained with AO. The cyanobacterium, ...

Increasing urbanization, industrialization and population growth in developing countries of the Third World will potentially increase the number of hypertrophic freshwater ecosystems unless suitable effluent and water quality standards are promulgated. Abstract The literature was reviewed to determine the direct temperature effects on photosynthetic capacity (Pmax), specific respiration rate (Rest), and growth rate of bloom‐forming cyanobacteria (Anabaena, Aphanizomenon, Microcystis, Oscillatoria) and to assess the importance of direct tern‐perature effects on cyanobacterial dominance in lakes. This analysis is supported by field studies of Microcystis aeruginosa in a hypertrophic lake. The literature and field data show that Pmax, Rest, and growth ...
