Static friction between two very clean oxide-free copper single crystal (001) planes is studied by applying a horizontal force in a direction parallel to either [100] or [110] to a sphere placed on the horizontal surface of a fixed cube of the same crystallographic orientation. Cross sections, longtudinal sections and layer sections of the lower crystals are examined with an optical microscope, electron microscope and microhardness tester to find the extent of the occurrence of phenomena such as deformation, ...

The basic structure of the shaft of the bacterial flagellum is described and an account is given of work on polymorphism to date. The flagellum of wild-type Salmonella SJ670 has a characteristic left-hand helical form from pH 8 to pH 6, but undergoes two sharp transitions to other definite forms as the pH is lowered. The first transition, to a tight coil, can be followed by flow-birefringence. The flow-birefringence curve shows no evidence of hysteresis. Other details of the polymorphism, including ...

Flagellar motility was studied in detergent-treated cells of Chlamydomonas with a dark-field microscope equipped with high-powered illumination. Flagella tended to split into two or three fibers in the presence of ATP and detergent. Some of split fibers showed oscillatory bending motion.

A beta-actinin-like protein was isolated from plasmodia of the slime mold. The chain weight was the same as that of actin (43,000), but the amino acid composition was significantly different. The action of plasmodium beta-actinin on actin was the same as that of beta-actinin from rabbit skeletal muscle: inhibition of the recombination of F-actin fragments; formation of Mg polymer; inhibition of interfilamental interaction of F-actin and retardation of depolymerization of F-actin. The only difference ...

Helical transformations of reconstituted Salmonella flagella were visualized by dark-field light microscopy. Flagella from SJ670 strain were lefthanded helices with a pitch of 2.3 μm at neutral pH. When, however, the pH of the solution was lowered to 4.7, they were discontinouously transformed into close-coils with a pitch of 0.5 μm and a diameter of 1.2 μm, and a further lowering of the pH converted these coiled flagella into so-called curly ones, righthanded helices with a pitch of 1.1 μm. The ...

Reconstituted Salmonella flagella were found by dark field light microscopy to undergo reversible transformations at both acidic pH (Kamiya & Asakura, 1976) and alkaline pH. When the pH was increased from 7 to 12·5, flagella from strain SJ670 were rapidly transformed from a left-handed helix with a pitch of 2·3 μm (normal type) to a left-handed close-coil (coiled type) or to two types of right-handed helices with a pitch of 1·0 or 0·9 μm (curly I and curly II types). These helical structures were similar ...

[3H]thymidine incorporated into replicating P2 DNA during a brief pulse is found mainly in two size classes of DNA chains: short chains of about 10 S, and long chains of one genome length (32 S). The pulse-labelled 10 S short chains hybridize predominantly with the L-strand of the phage DNA, whereas the pulse-labelled genome-length DNA chains hybridize preferentially with the H-strand. From the direction of replication, the polarity of transcription and the data on hybridization of pulse-labelled ...

ABSTRACT The plasmodium of Physarum polycephalum contained 15·3 mmol Ca/kg fresh weight of sample, 11·8 mmol Mg/kg, 24·5 mmol K/kg and 1·4 mmol Na/kg. When the plasmodium was starved of food, the Ca content increased gradually up to 71·9 mmol/kg during 5 days of starvation. The concentration of other elements changed only slightly. The endoplasm contained 23·0 mmol Ca/kg, 12-6 mmol Mg/kg, 26·6 mmol K/kg and 1·7 mmol Na/kg, but these contents changed only slightly during starvation. The Ca, Mg, K ...

We have found that several kinds of helical flagella from Salmonella and Escherichia become straight in the presence of 0·5 m-citric acid at pH values below 4·0, while the straight flagella from a mutant Salmonella (SJ814) are transformed into a helical shape under the same conditions. These transformations are reversible and transitional. Current models of bacterial flagella (Calladine, 1976,1978; Kamiya, 1976) predict that the family of distinct wave-forms should include two types of straight flagella, ...

Dark-field microscopy with a high-powered light source revealed that the outer doublet microtubules (DMTs) from sea urchin (Pseudocentrotus depressus and Hemicentrotus pulcherrimus) sperm flagella assume helically coiled configurations (Miki-Noumura, T., and R. Kamiya. 1976. Exp. Cell Res. 97: 451.). We report here that the DMTs change shape when the pH or Ca-ion concentration is changed. The DMTs assumed a left-handed helical shape with a diameter of 3.7 +/- 0.5 micron and a pitch of 2.8 +/- 0.7 micron at ...

Flagellar filaments from three strains of Salmonella undergo reversible structural transitions between discrete helical forms when the temperature is changed under appropriate aqueous conditions. These transitions can be quantified by viscometry of concentrated flagellar solutions. A transition resulting in an increase in the right-handed structural twist of the filament is always exothermic. The change in van't Hoff enthalpy accompanying the transformation ranged between 90 kcal and 250 kcal per ...

ABSTRACT In a dumbbell-shaped plasmodium of Physarum polycephalum showing active shuttle streaming Ca was precipitated with potassium pyroantimonate (K[Sb(OH)6]), and the distribution of Ca between the cytoplasm and cellular organelles, especially vacuoles, was examined by electron microscopy. The contracting half-mass, where many empty vacuoles were present, was rich in the small Ca precipitates located in the cytoplasm. The relaxing half-mass, where many Ca-containing vacuoles were present, was ...

Among 256 consecutive subjects so far studied in our laboratory, we found one subject (a 63-year-old female) whose very low density lipoprotein (VLDL) and high density lipoprotein (HDL) contained unusually high amounts of apolipoprotein C-III-0 among apolipoprotein C-III polymorphic forms. Identification of apolipoprotein C-III-0 was achieved by a combination of basic polyacrylamide gel electrophoresis, isoelectric focusing and sialidase treatment of plasma apolipoproteins. This unusual lipoprotein ...

A nonmotile mutant of Salmonella typhimurium, SJW1254, has very short flagella (less than 0.1 micron long) due to a mutation in the structural gene of flagellin (H2). When ammonium sulfate was added to the culture medium of SJW1254 grown to the late-log phase, a large amount of protein precipitated. Gel electrophoresis and immunodiffusion showed that more than 90% (wt/wt) of the precipitated protein was flagellin. The mutant flagellin appeared to be excreted in the monomeric form, in an amount comparable ...

Abstract The transition from primary dormancy to secondary dormancy was examined using upper cocklebur ( Xanthium pennsylvanicum Wallr.) seeds. The non‐after‐ripened seeds with primary dormancy responded to chilling, anoxia, KCN, and NaN 3 with an increase in germination. However, their maximal responses to these treatments only occurred after a period of water imbibition, probably a reflection of the increasing growth potential of the axial tissue which was accompanied by the increase in the capacities ...
