In limbs with combined shortening with angulation or malrotation, deformity may be quickly or slowly corrected before lengthening with external fixation. We examined a series of 35 patients with 40 limbs that underwent acute deformity correction and subsequent gradual lengthening. The average deformity corrected was 19 °, with subsequent average lengthening of 4.1 cm. Good radiographic callus formation was noted in 34 of the 40 segments studied. A cell-free system consisting of ribosomes and cytosol fraction, prepared from rat anterior pituitary tissue, was shown to be active in the incorporation of radioactive leucine into protein. The incorporation of radioactive amino acid resembled protein synthesis in its requirement for ribosomes, cytosol fraction, Mg2+ and an energy source. Inappropriate stimulation or defective negative regulation of the type I interferon response can lead to autoinflammation. In genetically uncharacterized cases of the type I interferonopathy Aicardi–Goutières syndrome, we identified biallelic mutations in LSM11 and RNU7-1, which encode components of the replication-dependent histone pre-mRNA–processing complex.
