The MAPK-interacting kinase (Mnk) family includes Mnk1 and Mnk2, which are phosphorylated and activated in response to extracellular stimuli. Mnk1 contributes to cellular responses by regulating messenger RNA (mRNA) translation, and mRNA translation influences platelet production and function. However, the role of Mnk1 in megakaryocytes and platelets has not previously been studied. The present study investigated Mnk1 in megakaryocytes and platelets using both pharmacological and genetic approaches. ...

Nearly 70% of adults in the US are currently overweight or obese. Despite such high prevalence, the impact of obesity on anti-tumor immunity and immunotherapy outcomes remains incompletely understood, particularly in patients with breast cancer. Here, we addressed these gaps in knowledge using two murine models of breast cancer combined with diet-induced obesity. We report that obesity increases CXCL1 concentrations in the mammary tumor microenvironment, driving CXCR2-mediated chemotaxis and accumulation ...

This study examined the psychometric structure of the Mental Health Inventory (MHI) in 433 cancer patients. Using structural equation modeling, confirmatory factor analyses (CFAs) were conducted. Next, exploratory factor analysis (EFA) was used to explore an alternative MHI factor structure with a randomly chosen subsample. Finally, CFAs were conducted on 6 MHI models with the second subsample. Convergent validity was examined by administering the Positive and Negative Affect Schedule (PANAS) and the ...

Substance P (SP) plays a central role in the transduction of second messenger signals from primary afferent nociceptive terminals to second-order neurons in the spinal cord. We have tested a recombinant engineered diphtheria toxin/SP fusion protein (DAB389SP) in acute and chronic pain models in the rat. DAB389SP binds to the SP receptor (SPR) and is internalized and kills SPR-expressing cells by blocking cellular protein synthesis. DAB389SP delivery was by intrathecal infusion, of varying duration, ...

The spinal cord is the site of many chronic, debilitating, neurological disorders that may be amenable to gene therapy. The present study, using quantitative and anatomical methods, examines the ability of replication deficient adenovirus to transfer a transcription cassette composed of the cytomegalovirus promoter driving the expression of the LacZ reporter gene (AdCMVβgal) to spinal-cord neurons. Rats were microinjected with AdCMVβgal into the spinal-cord parenchyma or subarachnoid space and sacrificed ...

MERGE provides a framework for thinking about climate change management proposals. The model is designed to be sufficiently flexible to be used to explore alternative views on a wide range of contentious issues, eg costs, damages, valuation and discounting. We begin with a description of the model's individual components and show how they fit together. We then provide an initial application to illustrate how the framework can be used in the assessment of alternative policy options. Given the level ...

In 27 patients with 74 colonic polyps we prospectively evaluated polyp-related risk factors and specific histopathological characteristics that might result in blood loss from these lesions. Each polyp was endoscopically bathed in saline which was then aspirated back into a syringe through a catheter and tested for presence of blood on a Hemoccult II slide. Forty-nine polyps were excised. We found the following: 1) A relatively small proportion of adenomas (∼11%) have a propensity to bleed. 2) Approximately ...

We present simple method to assess dental pain in the awake rat. Using a sensitive strain gauge we examined changes in bite strength and bite pattern in rats following dental injury. Rats with dental injury displayed a significant reduction in mean peak bite strength and an altered bite cluster pattern. Both changes in the dental injury rats were reversed by an analgesic dose of morphine, and this could be reversed with naloxone. These changes were not observed in naive control animals. Abstract Hepatitis B Virus (HBV) transgenic mice replicating the viral genome at high level but lacking expression of the small envelope protein (HBsAg) have been produced using a terminally redundant viral DNA construct (HBV 1.4). The generation of viable infectious progeny was dependent on sex and age of mice. Viral mRNA was abundant in liver and kidneys and at low levels in other organs of the mice. No viral particles or HBV envelope proteins could be detected in sera of mice. Despite expression ...

We present near-infrared light curves of supernova (SN) 2011fe in M101, including 34 epochs in H band starting 14 days before maximum brightness in the B band. The light curve data were obtained with the WIYN High-Resolution Infrared Camera. When the data are calibrated using templates of other Type Ia SNe, we derive an apparent H-band magnitude at the epoch of B-band maximum of 10.85 ± 0.04. This implies a distance modulus for M101 that ranges from 28.86 to 29.17 mag, depending on which absolute calibration ...
