The genetic mechanisms of human immunodeficiency virus type 1 (HIV-1) resistance to dideoxyinosine (ddI) in vivo have been described based on data from primary HIV-1 isolates. To better define the spectrum of HIV-1 reverse transcriptase (RT) changes occurring during ddI therapy, we determined the genotype and ddI susceptibility of the RT gene of HIV RNA isolated from the plasma of 23 patients who had received 1 to 2 years (mean, 87 +/- 16 weeks) of ddI monotherapy. Population-based sequencing of plasma ...

This paper describes the design and performance of the beam position detector being installed in the Fermilab superconducting Energy Doubler accelerator. The detector is a stripline pickup designed to operate at 4 degrees Kelvin and with beam intensities ranging from 108 to 1011 protons per RF bucket at 53 MHz. The detector design, signal amplitude, positional sensitivity, directivity, and longitudinal impedance are presented.

Graft versus host disease (GVHD) and graft rejection prophylaxis was with tacrolimus and mycophenolate mofetil. Thirteen of the 21 patients (62%) had mixed chimerism (≤≤90% donor cells) at day 60 and 11 (52%) of these patients had mixed chimerism which persisted until day 100. There is much ambiguity and variation in what the term cellular manufacturing refers to in both academia and industry. To help clarify the definition of cellular manufacturing a taxonomy for equipment groupings in batch environments is presented. The taxonomy categorizes equipment groupings on two continuous dimensions: the purpose for grouping equipment together into cells, and the amount of interdependence between the cells. Operational implications and insights obtained by mapping academic studies ...

Tuberculosis (TB) is the most common opportunistic infection and the leading cause of death in persons infected with human immunodeficiency virus (HIV) worldwide. Because HIV is spreading in regions with the highest rates of Mycobacterium tuberculosis infection, HIV is responsible for an increasing proportion of the world's cases of TB. However, advances in molecular biology, clinical practice, and public health policy during the past 5 years offer reasons for hope. Molecular methods have provided ...

Genetic research is increasingly turning to studies of sequence variation in genes encoding proteins of known structure and function. The principal question in these studies is whether sequence variation affects protein structure or function, and, for certain genes, whether sequence variation affects human health. The proliferation of published sequence data and the growth in the number of publications is a boon to this research, but also makes it difficult to keep track of what is known about a ...

Background: Dolutegravir (DTG)-based antiretroviral therapy (ART) rarely leads to virological failure (VF) and drug resistance in integrase strand transfer inhibitor (INSTI)-naïve persons living with HIV (PLWH). As a result, limited data are available on INSTI-associated drug resistance mutations (DRMs) selected by DTG-containing ART regimens. Methods: We reviewed studies published through July 2023 to identify those reporting emergent major INSTI-associated DRMs in INSTI-naïve PLWH receiving DTG and ...

Abstract Introduction HIV‐1 transmitted drug resistance (TDR) prevalence increased during the initial years of the antiretroviral therapy (ART) global scale‐up. Few studies have examined recent trends in TDR prevalence using published genetic sequences and described the characteristics of ART‐naïve persons from whom these published sequences have been obtained. Methods We identified 125 studies published between 2014 and 2019 for which HIV‐1 reverse transcriptase (RT) with or without protease from ≥ ...

The World Health Organization Antiretroviral Treatment Guidelines recommend phasing-out stavudine because of its risk of long-term toxicity. There are two mutational pathways of stavudine resistance with different implications for zidovudine and tenofovir cross-resistance, the primary candidates for replacing stavudine. However, because resistance testing is rarely available in resource-limited settings, it is critical to identify the cross-resistance patterns associated with first-line stavudine ...

This module measures beam off-axis position and intensity over a 60db dynamic range. For 20 or more contiguous 53MHz beam bunches, the unit will derive a position signal for 108 to 1011 protons per bucket with a position accuracy of ±0.5 mm. Also, the unit will measure the position of 1010 or more protons in a single isolated rf bunch. A beam intensity ...

Abstract Background The pharmacokinetic profiles of sufentanil available in the literature are conflicting because of methodologic differences. Length of sampling and assay sensitivity are key factors involved in accurately estimating the volumes of distribution, clearances, and elimination phase. The unit disposition function of increasing doses of sufentanil were investigated and the influence of dose administered on the linearity of pharmacokinetics was assessed.
