Eversa® Transform 2.0 has been launched to be used in free form, but its immobilization may improve its performance. This work aimed to optimize the immobilization of Eversa® Transform 2.0 by the crosslinked enzyme aggregates (CLEAs) technique, using almost all the available tools to improve its performance. Several variables in the CLEA preparation were optimized to improve the recovered activity, such as precipitant nature and crosslinker concentration. Moreover, some feeders were co-precipitated ...

Abstract Graft-versus-host disease (GVHD) is a serious complication of allogeneic bone marrow transplantation, and donor T cells are indispensable for GVHD. Current therapies have limited efficacy, selectivity, and high toxicities. We used a novel flow cytometry technique for the analysis of intracellular phosphorylation events in single cells in murine BMT models to identify and validate novel GVHD drug targets.1-7 This method circumvents the requirement for large numbers of purified cells, unlike ...

The target of rapamycin (TOR) signaling regulates the nucleocytoplasmic shuttling of transcription factors in yeast. Whether the mammalian counterpart of TOR (mTOR) also regulates nucleocytoplasmic shuttling is not known. Using a phospho-specific monoclonal antibody, we demonstrate that mTOR phosphorylates Ser(168,170) of endogenous NFATc4, which are conserved gate-keeping Ser residues that control NFAT subcellular distribution. The mTOR acts as a basal kinase during the resting state to maintain ...

Abstract We develop quantum computational tools to predict the 3D structure of proteins, one of the most important problems in current biochemical research. We explain how to combine recent deep learning advances with the well known technique of quantum walks applied to a Metropolis algorithm. The result, QFold, is a fully scalable hybrid quantum algorithm that, in contrast to previous quantum approaches, does not require a lattice model simplification and instead relies on the much more realistic ...

There is a pressing need to develop new rechargeable battery technologies that can offer higher energy storage, faster charging, and lower costs. Despite the success of existing methods for the simulation of battery materials, they can sometimes fall short of delivering accurate and reliable results. Quantum computing has been discussed as an avenue to overcome these issues, but only limited work has been done to outline how they may impact battery simulations. In this work, we provide a detailed ...

Smart homes allow optimized energy usage, allowing households to reduce electricity bills or even make profits. By 2020, 20% of all households in Europe will be expected to become smart homes. Although smart homes seem to be the future for homes, many customers have the perception that a transition from current homes to smart ones is unprofitable. Adopting a smart home concept requires investments for which the households desire a positive return. A question in this context is the following: for ...

The loss of canopy-forming seaweeds from urbanized coasts has intensified in response to warming seas and non-climatic pressures such as population growth and declining water quality. Surprisingly, there has been little information on the extent of historical losses in the South-western Atlantic, which limits our ability to place this large marine ecosystem in a global context. Here, we use meta-analysis to examine long-term (1969-2017) changes to the cover and biomass of Sargassum spp. and structurally ...

CT perfusion (CTP) and diffusion or perfusion MRI might assist patient selection for endovascular thrombectomy. We aimed to establish whether imaging assessments of irreversibly injured ischaemic core and potentially salvageable penumbra volumes were associated with functional outcome and whether they interacted with the treatment effect of endovascular thrombectomy on functional outcome. In this systematic review and meta-analysis, the HERMES collaboration pooled patient-level data from all randomised ...

Background Evidence regarding whether imaging can be used effectively to select patients for endovascular thrombectomy (EVT) is scarce. We aimed to investigate the association between baseline imaging features and safety and efficacy of EVT in acute ischaemic stroke caused by anterior large-vessel occlusion. Methods In this meta-analysis of individual patient-level data, the HERMES collaboration identified in PubMed seven randomised trials in endovascular stroke that compared EVT with standard medical ...

Reliable cell recovery and expansion are fundamental to the successful scale-up of chimeric antigen receptor (CAR) T cells or any therapeutic cell-manufacturing process. Here, we extend our previous work in whole blood by manufacturing a highly parallel deterministic lateral displacement (DLD) device incorporating diamond microposts and moving into processing, for the first time, apheresis blood products. This study demonstrates key metrics of cell recovery (80%) and platelet depletion (87%), and it ...

White metal alloys, known as Babbitt, are used for sliding in hydrodynamic bearings. The main objective of this study is to evaluate and compare the microstructure and mechanical and tribological properties of ASTM B23/2 alloy deposited by Arc Spray Process and Flame Spray, with coatings deposited by centrifugal and conventional casting processes. It was observed that the coatings deposited by thermal spraying processes had higher porosity and microstructural refinement than conventional samples. ...
