Environmental pollutants have recently emerged as potential risk factors for metabolic diseases, urging systematic investigation of pollutant effects on metabolic disease processes. To enable risk assessment of these so-called metabolic disruptors the use of stable, robust and well-defined cell based screening systems has recently been encouraged. Since beta-cell (dys)functionality is central in diabetes pathophysiology, the need to develop beta-cell based pollutant screening systems is evident. ...

Climate change is expected to shift the boreal biome northward through expansion at the northern and contraction at the southern boundary respectively. However, biome-scale evidence of such a shift is rare. Here, we used remotely-sensed tree cover data to quantify temporal changes across the North American boreal biome from 2000 to 2019. We reveal a strong north-south asymmetry in tree cover change, coupled with a range shrinkage of tree cover distributions. We found no evidence for tree cover expansion ...

Patients with chronic kidney disease (CKD) exhibit a high prevalence of neuropsychiatric alterations, including depression and behavioural changes. CKD is also associated with decreased physical activity not fully explained by co-morbidities. In patients without CKD, the brain-derived neurotropic factor (BDNF) as well as the endogenous NOS inhibitor asymmetric dimethylarginine (ADMA) had been suspected to be involved in major depression. The aim of our study was to examine the role of ADMA and BDNF ...

Asymmetric dimethylarginine (ADMA) is an endogenous inhibitor of nitric oxide synthases (NOS). Reducing inducible NOS activity in acute inflammation seems to be desirable. In vitro data show that ADMA increases in response to inflammatory mediators, yet the effect of acute inflammation in vivo is scarcely studied. The aim of the study was to evaluate ADMA plasma levels before, during, and after the acute (nonbacterial) inflammatory-like state. Plasma ADMA, l-arginine, C-reactive protein, and IL-6 were ...

To reduce the consequences of infectious disease outbreaks, the timely implementation of public health measures is crucial. Currently used early-warning systems are highly context-dependent and require a long phase of model building. A proposed solution to anticipate the onset or termination of an outbreak is the use of so-called resilience indicators. These indicators are based on the generic theory of critical slowing down and require only incidence time series. Here we assess the potential for ...

Abstract. Modeling studies have shown the importance of biogeophysical effects of deforestation on local climate conditions but have also highlighted the lack of agreement across different models. Recently, remote-sensing observations have been used to assess the contrast in albedo, evapotranspiration (ET), and land surface temperature (LST) between forest and nearby open land on a global scale. These observations provide an unprecedented opportunity to evaluate the ability of land surface models ...

Abstract As part of the REgional Carbon Cycle Assessment and Processes Phase 2 (RECCAP2) project, we developed a comprehensive African Greenhouse gases (GHG) budget covering 2000 to 2019 (RECCAP1 and RECCAP2 time periods), and assessed uncertainties and trends over time. We compared bottom‐up process‐based models, data‐driven remotely sensed products, and national GHG inventories with top‐down atmospheric inversions, accounting also for lateral fluxes. We incorporated emission estimates derived from ...

During the past decades managed forest ecosystems in Central Europe underwent vast changes, induced by extreme climate conditions and occasionally adverse forest management. Tree ring width patterns mirror these changes and thus have been widely examined as environmental archives and reliable empirical data sources in 'tree growth modelling'. Dendrochronologists often suppose linear co-variation among the covariates, variable independence and homoscedasticity. Conventionally, these assumptions were ...

Abstract Ecosystem regime shifts can have severe ecological and economic consequences, making it a top priority to understand how to make systems more resilient. Theory predicts that spatial connectivity and the local environment interact to shape resilience, but empirical studies are scarce. ABSTRACT Among the many concerns for biodiversity in the Anthropocene, recent reports of flying insect loss are particularly alarming, given their importance as pollinators, pest control agents, and as a food source. Few insect monitoring programmes cover the large spatial scales required to provide more generalizable estimates of insect responses to global change drivers. We ask how climate and surrounding habitat affect flying insect biomass using data from the first year of a new monitoring network ...

Extreme-ultraviolet to x-ray free-electron lasers (FELs) in operation for scientific applications are up to now single-user facilities. While most FELs generate around 100 photon pulses per second, FLASH at DESY can deliver almost two orders of magnitude more pulses in this time span due to its superconducting accelerator technology. This makes the facility a prime candidate to realize the next step in FELs—dividing the electron pulse trains into several FEL lines and delivering photon pulses to ...

Abstract According to the alternative prey hypothesis (APH), the temporal synchrony in population fluctuations of microtine rodents and other small herbivores in boreal areas is caused by generalist predators with numerical and functional response to microtines, leading to an increased predation of prey alternative to microtines in the low phase of the microtine population fluctuations. The boreal forest biome is warming four times faster than the global average. Changes so far are moderate, but time lags in responses may transiently maintain forest states which are no longer supported by current environmental conditions. Here, we explore whether tree cover dynamics hint at the state to which the biome may be shifting. Our results suggest that global boreal tree cover is heading for a unimodal state of open forest (30 to 50%), contrasting starkly to the bimodal distribution of dense ...

Abstract The catalog of cancer driver mutations in protein-coding genes has greatly expanded in the past decade. However, non-coding cancer driver mutations are less well-characterized and only a handful of recurrent non-coding mutations, most notably TERT promoter mutations, have been reported. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium, which aggregated whole genome sequencing data from 2658 cancer across 38 tumor types, we perform multi-faceted pathway ...

Abstract A key mutational process in cancer is structural variation, in which rearrangements delete, amplify or reorder genomic segments that range in size from kilobases to whole chromosomes 1–7 . Here we develop methods to group, classify and describe somatic structural variants, using data from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA), which aggregated whole-genome sequencing data from 2,658 ...

Abstract Cancers require telomere maintenance mechanisms for unlimited replicative potential. They achieve this through TERT activation or alternative telomere lengthening associated with ATRX or DAXX loss. Here, as part of the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium , we dissect whole-genome sequencing data of over 2500 matched tumor-control samples from 36 different tumor types aggregated within the ICGC/TCGA Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium to characterize ...

Abstract About half of all cancers have somatic integrations of retrotransposons. Here, to characterize their role in oncogenesis, we analyzed the patterns and mechanisms of somatic retrotransposition in 2,954 cancer genomes from 38 histological cancer subtypes within the framework of the Pan-Cancer Analysis of Whole Genomes (PCAWG) project. We identified 19,166 somatically acquired retrotransposition events, which affected 35% of samples and spanned a range of event types. Long interspersed nuclear ...
