Organs of rats treated with the drug A Y-9944 for 5 days showed a significant reduction in sphingomyelinase activity. Evidence is presented which suggests that the reduction is due to impaired enzyme synthesis.

This paper reconsiders Japan's fiscal sustainability. We investigate whether a simulation conducted under the political constraint imposed by a fiscal reaction function supports the official projection. First, we obtain Japan's fiscal reaction function by estimating the response of the primary surpluses to the past debt for a panel data set of 23 OECD countries. Next, we investigate the political feasibility of the official projection using our estimated reaction function. When the Cabinet Office ...

Thrombin formation was studied with the use of purified prothrombin, prothrombin complex, prethrombin, Ac-globulin, platelet factor 3, autoprothrombin C, autoprothrombin III (Factor X), and thrombin, With Auto-C alone, there was only a fractional yield of the potential thrombin from purified prothrombin complex or purified prothrombin. This low yield of thrombin was partly due to the simultaneous formation of prethrombin and autoprothrombin II-A, which is a competetitive inhibitor of Auto-C. Echis ...

Methods were developed for the assay and isolation of autoprothrombin II-A. The purified protein retarded blood coagulation in vitro as well as in vivo and induced fibrinolysis. It is tentatively concluded that the protein originated from prothrombin and possessed inhibitor-activator qualities respectively as anticoagulant and fibrinolytic agent. To obtain Auto II-A, concentrated prothrombin complex was digested with purified thrombin. Chromatography on a DEAE-cellulose column separated prethrombin, ...

Abstract Myelin purified from the central nervous system of Xenopus laevis contained the same major lipid and protein components as human myelin. However, some minor differences in the myelin proteins were noted. The Xenopus basic protein had a higher apparent mol wt. on sodium dodecyl sulfate gels than the corresponding mammalian protein. The absolute specific activity of 2′,3′‐cyclic nucleotide 3′‐phosphohydrolase in the Xenopus myelin was considerably higher than in mammals. There were differences ...

Pathologic cells of acute promyelocytic leukemia were studied as a potential trigger substance for disseminated intravascular coagulation syndrome (DIC). The pathologic cells had both procoagulant and fibrinolytic activity. When this substance was separated by Sephadex G-200 column chromatography, procoagulant activity and fibrinolytic activity were observed in the same peak. Procoagulant activity was proven by prothrombin conversion activity, and fibrinolytic activity was proven by plasminogen free ...

Lens cells of congenital mouse cataract (Nakano and Fraser strains) and galactose-fed rats were studied by scanning electron microscopy. Similarly lens cells of normal young mice and rats were examined as controls. Normal lenses of young rodents consist of lens fibers in all maturation stages which have been demonstrated in humans and in monkeys. Lens cells in the congenitally cataractous lenses are irregular in size and shape in the earlier stage of the cataract formation. Swelling of the lens cell ...

Prethrombin 1 was found in the blood of patients who have the disseminated intravascular coagulation syndrome (DIC). Prothrombin assay method using venom is useful for detecting prothrombin derivatives such as prethrombin 1 (1) in DIC. This prethrombin is produced from prothrombin by the proteolytic effect of thrombin and is difficult to convert to thrombin by physiologic procoagulants. This fact is demonstrated by a bioassay of the interaction between purified prothrombin and purified thrombin and ...

Abstract Two cases of von Willebrand disease (VWD) that revealed an increased ristocetin‐induced platelet aggregation (RIPA) and a qualitative abnormality of the factor VIII protein are reported. The threshold concentration of ristocetin giving a 30% increase in light transmission was 0.5 mg/ml in the proband and 0.4 mg/ml in her father (normal: 1.16 ± SD 0.18 mg/ml) although both patients showed reduced plasma von Willebrand factor activity (VIIIR:WF). In both patients, the amount of factor VIII related ...

Human placental acid sphingomyelinase was highly purified in the presence of Triton X-100. DEAE-Sephacel chromatography and chromatofocusing were the most effective steps in the purification procedure. Enzyme purification was 380,000 nmol/mg protein/h. Characterization and radioiodination were carried out with the chromatofocusing fraction containing highly purified enzyme. The purified enzyme contained no activity of eleven other lysosomal hydrolases but hydrolyzed bis- p -nitrophenyl phosphate ...

The oral administration of heparin alone or entrapped in liposomes was investigated. The prolongation of clotting time (a-PTT) was observed after administration of heparin and attributed to an increase of heparin activity in the blood as a result of effective absorption from the intestine. The increase of heparin activity was promoted by entrapping heparin in liposomes, but not by mere addition of empty liposomes.

Tohkaku-johkitoh and keishi-bukuryo-gan showed remarkable inhibitory effects on fibrinolysis, coagulation and platelet aggregation.
