A new type of electron spin−echo envelope modulation (ESEEM) was observed in a three-pulse stimulated echo experiment on nitroxides in a molecular glass. With fixed interval T between the second and the third pulses and variable interval τ between the first and the second ones, it appears in the frequency domain as a broad peak around ∼5 MHz. It is attributed to a sudden change of the EPR resonance frequency due to reorientation of the methyl groups of the nitroxide during the T interval. The rate ...

Abstract We conducted a detailed kinetic study of the reaction of the vitamin B 12 analog diaquacobinamide [(H 2 O) 2 Cbi(III)] with hydrogen sulfide in water from pH 3 to 11. The reaction proceeds in three steps: (1) the formation of three different complexes between cobinamide and hydrogen sulfide, viz., (HO – )(HS – )Cbi(III), (H 2 O)(HS – )Cbi(III), and (HS – ) 2 Cbi(III); (2) inner‐sphere electron transfer (ISET) in the two complexes with one coordinated HS – ligand to form the reduced cobinamide complex [(H)S]Cbi(II); ...

We studied the kinetics of reactions of cob(I)alamin and cob(I)inamide with thiosulfate, sulfite, and dithionite by UV-Visible (UV-Vis) and stopped-flow spectroscopy. We found that the two Co(I) species were oxidized by these sulfur-containing compounds to Co(II) forms: oxidation by excess thiosulfate leads to penta-coordinate complexes and oxidation by excess sulfite or dithionite leads to hexa-coordinate Co(II)–SO2− complexes. The net scheme involves transfer of three electrons in the case of oxidation ...

The interaction of Co(III) and Co(II) cobalamin (Cbl) and cobinamide (Cbi) with thiocyanate was examined with UV–vis and EPR spectra. S/N-linkage isomerism was explored on Co(III) and Co(II) Cbl and Cbi models using density functional theory (DFT; BP86, B3LYP). Performed calculations suggest the prevalence of isothiocyanato isomers over thiocyanato complexes on both Co(III) and Co(II) centers. The formation of Cbl(II) complex with thiocyanate was observed at high ligand concentrations which was proposed ...

A detailed study of the oxidation of L-ascorbic acid by dioxygen and nitrite in water at pH 5.8 and 7.0, catalyzed by the octasulfophenyltetrapyrazinoporphyrazine complex of cobalt(II), was carried out using conventional spectrophotometric, low-temperature and high-pressure stopped-flow techniques. The Co(II) complex activates L-ascorbic acid through an intramolecular one-electron oxidation step that involves the reduction of the octasulfophenyltetrapyrazinoporphyrazine. The reaction rate strongly ...

Evidence for the formation of a unique, six-coordinate cobalamin(II) complex with the anion-radical SO2− during the reduction of aquacobalamin(III) by sodium dithionite, was obtained from spectrophotometric and EPR measurements. The pKa value of the weakly coordinated dimethylbenzimidazole group was found to be 4.8 ± 0.1 at 25 °C.

A comprehensive review on redox reactions of Co(III), Co(II) and Co(I) forms of cobalamins (vitamin B12) and their derivatives (cobinamide, cobyrinates) is presented. An influence of coordination environment of cobalt ion on reactivity of corresponding species is discussed. Mechanisms of reduction of Co(III) and Co(II) species as well as oxidation of Co(II) and Co(I) complexes are given in terms of their in vivo and catalytic implications.

The reaction of cyanocobalamin (CNCbl) with sodium hydroxymethanesulfinate (HMS) was studied over a wide range of pH (4–11) under aerobic conditions. CNCbl is destroyed in the presence of HMS in aqueous solution to form uncolored substances. The accumulation of stable yellow corrinoids (SYCs) preceded these changes at pH [Formula: see text] 8. The major stable yellow corrinoid is (15R)-Co[Formula: see text], Coß — dicyano-13-dehydro-15-hydro-l5-hydroxycob(III)alamin. The yield of this SYC is 25%, and the ...

This work reports the reactivity of cob(II)alamin (Cbl(II)) toward reduction of dehydroascorbic (DHA) to ascorbic acid (AA) mediated by sulfur-containing compounds such as glutathione (GSH) and thiocyanate. The reaction supported by GSH proceeded more efficiently than with SCN−. Our findings demonstrate new aspects of interactions between vitamins B12 and C. It has been accepted that simultaneous presence of these vitamins results in their decomposition (viz., irreversible modification of the corrin ...

Emerging viruses threaten global health, but few experimental models can characterize the virus and host factors necessary for within- and cross-species transmission. Here, we leverage a model whereby pet store mice or rats-which harbor natural rodent pathogens-are cohoused with laboratory mice. This "dirty" mouse model offers a platform for studying acute transmission of viruses between and within hosts via natural mechanisms. We identified numerous viruses and other microbial species that transmit ...

For dipalmitoylphosphatidylcholine (DPPC) lipid/water bilayers, a detailed temperature dependence of the Raman scattering spectra at the spectral range of the CH2-stretching modes was investigated. Below 150 K the ratio of intensities of the 2880 cm−1 antisymmetric vibration line and the 2850 cm−1 symmetric one was found to be nearly temperature-independent. Between 150 and 230 K it decreases slightly as temperature increases; and above 230 K it decreases remarkably. This decrease is accompanied ...

Continuous wave (cw) electron paramagnetic resonance (EPR) and echo-detected (ED) EPR were applied to study molecular motions of nitroxide spin probes in glassy glycerol and o-terphenyl. A linear decrease with increasing temperature of the total splitting in the cw EPR line shape was observed at low temperatures in both solvents. Above some temperature points the temperature dependencies become sharper. Within the model of molecular librations, this behavior is in qualitative and quantitative agreement ...

A comparative study of anisotropic relaxation in two-pulse primary and three-pulse stimulated electron spin echo decays provides a direct way to distinguish fast (correlation time tau(c) 10(-6) s) motions. Anisotropic relaxation is detected as a difference of the decay rates for different resonance field positions in anisotropic electron paramagnetic resonance spectra. For fast motion anisotropic relaxation influences the primary echo decay and does not influence the stimulated echo decay. For slow ...

Sulfoxylate SO 2 H − ( SO 2 2− ), a strong reducing agent readily produced by hydrolysis of thiourea dioxide, reacts with ferric myoglobin ( Mb ) to reversibly produce Fe (II)- Mb , starting from either aerobic or anaerobic conditions. Exposure of Fe (II)- Mb to excess sulfoxylate further produces Fe (II)- CO - Mb . Fe (II)- Mb can be regenerated by reoxidation with ferricyanide at this stage; hemin, rubredoxin and cytochrome c show a similar reactivity towards sulfoxylate. The source of CO is not the protein moiety, ...
