The metabolism of [24-14C]lithocholic acid was studied in the microsomal fraction of fetal human liver homogenates. Fetal livers were obtained at legal abortions performed between wk 14 and 24. Product formation was analyzed by thin-layer chromatography, liquid chromatography, and gas chromatography-mass spectrometry. From wk 14 lithocholic acid was hydroxylated in several positions. Hyodeoxycholic acid (3 alpha,6 alpha-dihydroxy-5 beta-cholanoic) and 1 beta,3 alpha-dihydroxy-5 beta-cholanoic acid ...

We investigate the applicability of effective medium models for describing the effects of surface roughness on ellipsometric data. The ellipsometric parameters of a model surface composed of truncated spherical bumps on a substrate were calculated and compared to the Bruggeman effective medium theory. It is possible to fit the model calculations with the Bruggeman theory. However, the thickness and volume fraction of the effective medium layer describing the surface roughness do not correspond to ...

Fluxes of trace metals across the sediment-water interface were measured in situ at 6 m depth in Gullmarsfjorden, Sweden, using diver-operated stirred benthic flux-chambers. These were equipped so that dissolved oxygen and pH could be maintained near ambient seawater values (regulated chamber) or be allowed to change in response to benthic respiration (unregulated chamber). In the regulated chamber, Cd, Cu, Ni, and Zn were released from the sediment at constant rates both during a winter experiment ( ...

Aggregated distributions of macroparasites within their host populations are characteristic of most natural and experimental infections. We designed this study to measure the amount of variation that is attributable to host genetic factors in a pig–helminth system. In total, 195 piglets were produced after artificial insemination of 19 sows (Danish Landrace–Yorkshire crossbreds) with semen selected from 13 individual Duroc boars (1 or 2 sows per boar; mean litter size: 10.3; 5–14 piglets per litter). ...

Background: In Sweden, patients with diabetes mellitus frequently receive short-term (<3 months) continuous glucose monitoring (CGM) to study glucose patterns or long-term CGM to treat poor glycemic control or severe hypoglycemia. The effects of CGM on glycemic control in clinical practice in relation to indication and duration of use has not been completely studied. Methods: Patients with diabetes, among which 99% were diagnosed as type 1, receiving CGM at 10 outpatient clinics in Sweden were studied ...

Skin is easily accessible for transdermal drug delivery and also attractive for biomarker sampling. These applications are strongly influenced by hydration where elevated hydration generally leads to increased skin permeability. Thus, favorable transdermal delivery and extraction conditions can be easily obtained by exploiting elevated skin hydration. Here, we provide a detailed in vivo and in vitro investigation of the skin hydration dynamics using three techniques based on electrical impedance ...

Reliable quantitative vegetation reconstructions for Europe during the Holocene are crucial to improving our understanding of landscape dynamics, making it possible to assess the past effects of environmental variables and land-use change on ecosystems and biodiversity, and mitigating their effects in the future. We present here the most spatially extensive and temporally continuous pollen-based reconstructions of plant cover in Europe (at a spatial resolution of 1° × 1°) over the Holocene (last 11. ...

Hepatocyte growth factor (HGF) is a cytokine produced by myeloma cells. We examined serum HGF levels in a population of young myeloma patients (median age 52 years) treated with high-dose chemotherapy. Sera from 128 myeloma patients at diagnosis and serial samples from 16 patients were analysed. Compared with 62 healthy controls, HGF was elevated at diagnosis in 25% of patients (median 0.48 and 1.08 ng/ml respectively; P < 0.0001). The 95 patients who completed therapy were analysed for the impact of ...

Proteasome inhibitors have demonstrated that targeting protein degradation is effective therapy in multiple myeloma (MM). Here we show that deubiquitylating enzymes (DUBs) USP14 and UCHL5 are more highly expressed in MM cells than in normal plasma cells. USP14 and UCHL5 short interfering RNA knockdown decreases MM cell viability. A novel 19S regulatory particle inhibitor b-AP15 selectively blocks deubiquitylating activity of USP14 and UCHL5 without inhibiting proteasome activity. b-AP15 decreases ...

Metronidazole is traditionally dosed every 6-8 hours even though in adults it has a long half-life, concentration-dependent killing, and 3-hour postantibiotic effect. Based on this logic, some pediatric hospitals adopted once-daily dosing for appendicitis, despite limited pharmacokinetics-pharmacodynamics (PK/PD) in children. We studied pediatric patients with appendicitis given metronidazole once daily to determine whether this dosing would meet target area under the curve (AUC)/minimum inhibitory ...

AbstractVanadium bromoperoxidase (VBrPO) from Corallina officinalis was demonstrated to be a potentially useful catalyst for the preparation of (S)-sulfoxides of high enantiomeric purity. No loss in enantioselectivity or yield was observed upon a thirtyfoid increase of the reaction scale in the oxidation of 2-methylsulfanyl-benzoic acid (2). The catalytic activity of VBrPO in asymmetric sulfoxidation was demonstrated to be dependent on the vanadium content in the active site. The enzyme was found to ...

The genotypes of both host and parasite may influence the outcome of parasitic infections, but few attempts have been made to quantify the effect of parasite genotype on macroparasite infections of socio-economic importance. We examined variation in particular traits during the infection in pigs with the parasitic nematode Ascaris suum. We infected 26 pigs with mixtures of equal proportions of embryonated eggs from 4 single female worms each with a unique mtDNA haplotype--the eggs from each female ...

A simple, one‐dimensional, transport‐reaction model of oxygen uptake by sediments during incubation experiments assumes zero‐order reaction kinetics for oxygen removal in the pore water of sediments and takes the apparent diffusive limitation posed by the benthic boundary layer into account. Model calculations are in excellent agreement with experimental data obtained during in situ benthic flux chamber experiments in Gullmarsfjorden, Sweden, during fall (water temp, 10°C) and winter (−1°C). The boundary ...

On the assumption that lack of oxygen will cause the activity of benthic infauna to cease and thereby limit solute transport to that accomplished by molecular diffusion, we have measured the flux of dissolved silicate across the sediment‐water interface of sediments incubated in situ under parallel benthic flux chambers. In one chamber the oxygen concentration was allowed to go to zero, and in the other the oxygen and pH remained at ambient values. As long as oxygen was present in the chambers, ...

The porewater of a sediment core taken at 6 m depth in Gullmarsfjorden, Sweden, was enriched in Fe, Mn, Co, and phosphate compared to the overlying bottom water. Yet, in situ measurements with a benthic flux-chamber, in which dissolved oxygen and pH were maintained near ambient values (regulated flux-chamber), showed that the sediment did not release any of these ions but instead removed Co, Mn, and Fe from the overlying water. In a parallel experiment, where dissolved oxygen and pH were not maintained ...

Falls among older adults are most frequently caused by slips and trips and can have devastating consequences. Perturbation-based balance training (PBT) have recently shown promising fall preventive effects after even small training dosages. However, the fall preventive effects of PBT delivered on a treadmill are still unknown. Therefore, this parallel-group randomised controlled trial aims to quantify the effects of a four-session treadmill-PBT training intervention on falls compared with treadmill ...

The suitability of total carbonate production instead of oxygen consumption as a measure of benthic respiration has been investigated. In situ fluxes of total carbonate, oxygen, calcium, total alkalinity, nutrients, and sulfide across the sediment‐water interface were measured in diver‐operated benthic flux chambers. Two chambers were run in parallel to test the influence of oxygen and pH levels on total carbonate production. In one, oxygen and pH were kept constant near ambient levels; in the other, ...
