Protein lysine methyltransferases are important regulators of epigenetic signaling. These enzymes catalyze the transfer of donor methyl groups from S-adenosylmethionine to specific acceptor lysines on histones, leading to changes in chromatin structure and transcriptional regulation. These enzymes also methylate nonhistone protein substrates, revealing an additional mechanism to regulate cellular physiology. The oncogenic protein SMYD2 represses the functional activities of the tumor suppressor ...

IL-12 and TNF-alpha production by dendritic cells (DCs) is a critical step in the initiation of local inflammation and adaptive immune responses. We show in this study that a small molecule immune response modifier that is a Toll-like receptor 7 (TLR7) agonist induces IL-12 and TNF-alpha production from murine CD11c(+)CD11b(+)CD8(-) DCs, a subset not previously known for this activity. Stimulation of these DCs through TLR7 in vivo induces significant cytokine production even 12 h after initial stimulation, ...

In England and Wales, Initial Teacher Education depends upon a legal and contractual partnership between schools and higher education institutions in which teacher mentors are often trained to support student teacher development. This paper investigates the much less formal kind of partnership model that has evolved in Scotland, using one university's PGCE (Secondary) programme as a case study. Our research shows that the partnership is based on goodwill and relationships built up over time between ...

Codeine can be determined by a Flow injection procedure based on the chemiluminescence generated by reaction with potassium permanganate (6 × 10–4 mol l–1) in the presence of 0.5 mol l–1 polyphosphoric acid. The limit of detection is 8 pmol (3 × 10–7 mol l–1 in a 25 µl sample), the relative standard deviation for the determination of 5 × 10–4 mol l–1 codeine is 1.4%(n= 10).

<p>This paper explores the contribution of social capital on the rural household food security. Social capital is the ability of community actors to secure benefits by virtue of membership in social networks or other structures. In the past decade, consensus has emerged among scholars and practitioners of development that social capital can contribute significantly to the alleviation of poverty. Food insecurity is an indicator of poverty. This paper therefore takes this view by investigating ...

SET and MYND domain-containing protein 2 (SMYD2) is a protein lysine methyltransferase that catalyzes the transfer of methyl groups from S-adenosylmethionine (AdoMet) to acceptor lysine residues on histones and other proteins. To understand the kinetic mechanism and the function of individual domains, human SMYD2 was overexpressed, purified, and characterized. Substrate specificity and product analysis studies established SMYD2 as a monomethyltransferase that prefers nonmethylated p53 peptide substrate. ...

The development of a novel method for absolute quantification of proteins based on isotope-coded affinity tagging using ICAT reagents is described. The method exploits synthetic peptide standards to determine protein content at the femtomole level in biological samples. The approach is generally applicable to any subset of proteins, but is particularly appropriate for quantitative analysis of multiple, closely related isoforms, and for hydrophobic proteins that are poorly represented in 2-D gels. ...

Biodiversity supports a range of ecosystems services that are of fundamental importance to people in poor countries. Economic valuation of biodiversity is important for the development of policies that protect biodiversity and alleviate poverty. This paper provides an evaluation of monetary and non-monetary techniques for assessing the value of biodiversity to people in least developed countries (LDCs). Specifically, research questions include: To what extent have monetary and non-monetary techniques ...

Toll-like receptors are important in the activation of innate immunity, and CD40 is a molecule critical for many T and B cell responses. Whereas agonists for either pathway have been used as vaccine adjuvants, we show that a combination of Toll-like receptor (TLR)7 and CD40 agonists synergize to stimulate CD8+ T cell responses 10–20-fold greater than the use of either agonist alone. Antigen-specific CD8+ T cells elicited from combination CD40/TLR7 treatment demonstrated both lytic activities and ...

Context. Brown dwarfs are poorly understood transition objects between stars and planets, with several competing mechanisms having been proposed for their formation. Mass measurements are generally difficult for isolated objects but also for brown dwarfs orbiting low-mass stars, which are often too faint for spectroscopic follow-up. Aims. Microlensing provides an alternative tool for the discovery and investigation of such faint systems. Here we present the analysis of the microlensing event OGLE- ...

Monetary valuation of ecosystem services enables more accurate accounting of the environmental costs and benefits of policies, but this has rarely been applied in developing countries. In such contexts, there are particular methodological and epistemological challenges that require novel valuation methodologies. This paper introduces a new participatory, deliberative choice experiment approach conducted in the Solomon Islands. The research aimed to determine the value people placed on ecosystem ...

Abstract A polymorphism in coagulation factor V, factor V Leiden (FVL), is the major known genetic risk factor for thrombosis in humans. Approximately 10% of mutation carriers experience clinically significant thrombosis in their lifetime. In a small subset of patients, thrombosis is associated with coinheritance of other prothrombotic gene mutations. However, the potential contribution of additional genetic risk factors in the majority of patients remains unknown. To gain insight into the molecular ...
