The objectives of this study were to identify tissue-specific differentially methylated regions (T-DMR's) in the folate transport genes in placental tissue compared with leukocytes, and from placental tissues obtained from normal infants or with neural tube defects (NTDs). Using pyrosequencing, we developed methylation assays for the CpG islands (CGIs) and the CGI shore regions of the folate receptor α (FOLR1), proton-coupled folate transporter (PCFT) and reduced folate carrier 1 (RFC1) genes. The T-DMRs ...

There is growing evidence that folate status and variation in folate-metabolizing genes are involved in female reproductive functions. This study evaluated the influence of maternal blood folate, vitamin B12, homocysteine and 10 folate pathway gene variants on IVF outcome. Also, the prevalence of these polymorphisms was compared in 439 female IVF patients and 225 fertile controls. MTHFR 677 CT heterozygotes had a higher proportion of good-quality embryos and an increased chance of pregnancy. MTHFR ...

Solute carrier family 25A member 43 (SLC25A43) gene is a putative tumor suppressor gene that undergoes loss of heterozygosity (LOH) in human epidermal growth factor receptor 2 (HER2) positive breast cancer. Also, knockdown of SLC25A43 in cell lines influences cell turnover and metabolism. Absence of mutations in this gene in breast cancers prompted us to study methylation as an alternate mechanism for gene inactivation of this X encoded gene. Quantification of CpG site methylation using pyrosequencing ...

During the last decade, graphene foam emerged as a promising high porosity 3-dimensional (3D) structure for various applications. More specifically, it has attracted significant interest as a solution for thermal management in electronics. In this study, we investigate the possibility to use such porous materials as a heat sink and a container for a phase change material (PCM). Graphene foam (GF) was produced using chemical vapor deposition (CVD) process and attached to a thermal test chip using sintered ...

Thermal treatment of carbon nanotubes (CNTs) can significantly improve their mechanical, electrical and thermal properties due to reduced defects and increased crystallinity. In this work we investigate the effect of annealing at 3000 °C of vertically aligned CNT arrays synthesized by chemical vapor deposition (CVD) on graphite. Raman measurements show a drastically reduced amount of defects and, together with transmission electron microscope (TEM) diffraction measurements, an increased average crystallite ...

A methionine‐containing tryptic peptide from acetamidinated human carbonic anhydrase B has been isolated. This peptide has been shown to bridge a previously known carboxyl‐terminal fragment prepared by CNBr degradation with the rest of the enzyme molecule. Sequence studies on the methionine‐containing peptide have extended the previous knowledge of the carboxyl‐terminal sequence of the enzyme. Corresponding bridge peptides from human carbonic anhydrase C and acetamidinated bovine carbonic anhydrase ...

Chitosanases have received much attention because of their wide range of applications. Although most fungal chitosanases use sugar as their major carbon source, in the present work, a chitosanase was induced from a squid pen powder (SPP)-containing Penicillium janthinellum D4 medium and purified by ammonium sulphate precipitation and combined column chromatography. The purified D4 chitosanase exhibited optimum activity at pH 7–9, 60 °C and was stable at pH 7–11, 25–50 °C. The D4 chitosanase that was ...

A new purification method for Cl̄-esterase inhibitor is described, which is essentially a three-step procedure: precipitation with poly(ethylene glycol), chromatography on DEAE-cellulose and hydrophobic interaction chromatography on hexyl-Sepharose. The final product is a single-chain glycoprotein with a molecular weight of about 100000 and NH2-terminal asparagine. The molecule is fully active as judged by complex formation with Cl̄s. Two of its three disulphide bridges can be easily reduced and ...

The properties of the one-chain and two-chain forms of the porcine tissue plasminogen activator have been compared with respect to their amidolytic activities against a low molecular weight synthetic substrate, and their inhibition by diisopropylphosphofluoridate, α2-antiplasmin and C1-esterase inhibitor. The two-chain form is the more efficient catalyst, and is more rapidly inhibited by diisopropylphosphofluoridate and α2-antiplasmin. Physiological implications are discussed.

A method for the specific determination of a form of alpha 2-antiplasmin which does not interact with the lysine-binding sites in plasminogen (NPB-AP) has been elaborated. Basically, the method is an electroimmnoassay utilizing an intermediate gel, which adsorbs the plasminogen-binding form of alpha 2-antiplasmin (PB-AP). The plasma concentration of NPB-AP in healthy individuals was determined as 0.40 mumol/1 +/- 0.14 (SD), constituting 35% +/- 11 (SD) of the total plasma alpha 2-antiplasmin concentration. ...

Summary Utilizing the immunoglobulin fraction from a goat antiserum against human uterine tissue plasminogen activator, an enzyme- linked immunoassay for tissue-type plasminogen activator in human plasma has been developed. With the new method, the concentration of t-PA in normal human acidified plasma is found to be 4.0 ± 1.8 (SD) ng/ml. It increases to 12 ng/ml after a tomiquet test, and to 14 ng/ml after strenous physical exercise. In a group of patients with idiopathic thromboembolic disease, the ...

The turnover of human tissue‐type plasminogen activator (t‐PA), purified from cell culture fluids and radiolabeled with 125 I, was studied in 2 healthy males. After injection, the plasma radioactivity initially disappeared with a half‐life of about 3 min, but after 30 min reached a plateau level which persisted for several hours. The radioactive material had then already been converted to forms soluble in 10% trichloroacetic acid (TCA). Surface counting rates indicated a rapid uptake in the liver ...
