Abstract The mechanisms for the control of ribosomal protein synthesis have been characterized in detail in Eukarya and in Bacteria. In Archaea, only the regulation of the MvaL1 operon (encoding ribosomal proteins MvaL1, MvaL10, and MvaL12) of the mesophilic Methanococcus vannielii has been extensively investigated. As in Bacteria, regulation takes place at the level of translation. The regulator protein MvaL1 binds preferentially to its binding site on the 23S rRNA, and, when in excess, binds to ...

In Bacteria and Archaea (formerly Archaebacteria) ribosomal protein L1 has a dual function, as a primary rRNA‐binding protein and as a translational repressor which binds to its own mRNA. The L1‐binding site on the mRNA exhibits high similarity in both sequence and secondary structure to the binding site for L1 on the 23 S rRNA. A sensitive membrane‐filter‐binding assay has been used to examine the interactions between ribosomal L1 proteins from different archaeal and bacterial species, and 23S rRNA ...

As in Escherichia coli , regulation takes place at the level of translation. MvaL1, the homologue of the regulatory protein L1 encoded by the L11 operon of E . coli , was shown to be an autoregulator of the MvaL1 operon. The regulatory MvaL1 ...

An Escherichia coli coupled in vitro transcription-translation system has been modified to allow efficient expression of genes under the control of a T7 promoter. We describe both the characterization and use of two S30 crude extracts prepared from E. coli, namely S30 BL21(DE3) (containing endogenous T7 RNA polymerase) and S30 BL21 (supplemented with exogenous T7 RNA polymerase). Since transcription by the highly active T7 RNA polymerase is known to overload the translational machinery of E. coli, ...

The ribosomal protein MvaS7 from the methanogenic archaeon Methanococcus vannielii is a protein of 188 amino acids, i.e., it is 42 amino acids longer than previously suggested. The triplet TTG serves as a start codon. The methanogenic translation initiation region that includes the rare TTG start codon is recognized in Escherichia coli.

The mechanisms for regulation of ribosomal gene expression have been characterized in eukaryotes and eubacteria, but not yet in archaebacteria. We have studied the regulation of the synthesis of ribosomal proteins MvaL1, MvaL10, and MvaL12, encoded by the MvaL1 operon of Methanococcus vannielii, a methanogenic archaebacterium. MvaL1, the homolog of the regulatory protein L1 encoded by the L11 operon of Escherichia coli, was shown to be an autoregulator of the MvaL1 operon. As in E. coli, regulation ...

Abstract. A large number of radiometers are traceable to the World Standard Group (WSG) for shortwave radiation and the interim World Infrared Standard Group (WISG) for longwave radiation, hosted by the Physikalisch-Meteorologisches Observatorium Davos/World Radiation Centre (PMOD/WRC, Davos, Switzerland). The WSG and WISG have recently been found to over- and underestimate radiation values, respectively (Fehlmann et al., 2012; Gröbner et al., 2014), although research is still ongoing. In view of a ...

The relationship between dietary protein intake and uric acid metabolism has been studied by many workers, such as Taylor and Rose (1914), Leopold et al. (1925), Bien et al. (1953), Waslien et al. (1968), and Bowering et al. (1969). However, this relationship was still unsettled as far as serum uric acid concentration is concerned. We therefore investigated the influence of a high dietary protein intake as compared to an average intake on serum and urinary uric acid.

Ten days after a healthy person has consumed a purine-free isoenergetic diet, the excretion of (endogenous) uric acid reaches a minimum and remains constant thereafter. Purines are absorbed in different degrees from various biochemical sources added to such a diet--little from DNA, nearly all from ribomononucleotides--presumably owing to different rates of hydrolysis. There is always a linear relationship between dietary purine content and uric acid excretion, independent of the source of purine. ...

The administration of allopurinol or oxipurinol leads to an apparent increase in the specific activity of orotate phosphoribosyltransferase and orotidine 5'-phosphate decarboxylase in man. In the present study, we have shown that human orotate phosphoribosyltransferase and orotidine 5'-phosphate decarboxylase exist in a complex as three different molecular species with molecular weights of 55,000, 80,000 and 113,000. The larger forms of the complex are more stable than the small form. In the presence ...

Orotic aciduria is a rare autosomal recessive disease in man due to a deficiency of orotate phosphoribosyltransferase (EC 2.4.2.10; orotidine-5'-phosphate:pyrophosphate phosphoribosyltransferase) and orotidine-5'-phosphate decarboxylase (EC 4.1.1.23; orotidine-5'-phosphate carboxy-lyase). We have compared certain physicochemical properties of orotidine-5'-phosphate decarboxylase from normal and mutant fibroblasts grown under identical conditions. Orotidine-5'-phosphate decarboxylase from homozygous ...

In recent years a number of specific concepts have been developed regarding the regulation of purine biosynthesis in man. At the present time, the most important factors involved in the control of this pathway appear to be the intracellular concentration of 5-phosphoribosyl-1-pyrophosphate (PP-ribose-P) and purine nucleotides. In addition, a number of enzymatic abnormalities, which may lead to an accelerated rate of purine biosynthesis de novo and hyperuricemia, have now been described. These include ...

Allopurinol-induced orotaciduria is reduced by dietary ribonucleic acid (RNA), RNA hydrolysate and different nucleotides. These findings are compatible with feedback regulation of pyrimidine biosynthesis by dietary nucleotides. Serum uric acid and urinary uric acid excretion on a purine-free isoenergetic diet reach a minimum after about 10 days and remain constant thereafter. When purines from different biochemical sources are added to such a diet there is always a linear relationship between dietary ...

Abstract The fluid‐filled stomach allows visualization of the gastric wall on real‐time ultrasonic examination. In four cases with small focal gastric wall lesions (two cases with hyperplastic polyps, one with pseudopolypoid lesions, and one with polypoid infiltration of the gastric mucosa by maligant lymphoma), typical ultrasonic pictures were demonstrated. The polypoid lesions were located on the anterior and posterior gastric walls and at the gastric fundus, respectively. All who use the gastric ...

The spread of Gram-negative bacteria with plasmid-borne extended-spectrum beta-lactamases (ESBLs) has become a worldwide problem. This study analysed a total of 366 ESBL-producing Enterobacteriaceae strains isolated from non-selected patient specimens at the university hospital of Tübingen in the period January 2003 to December 2007. Although the overall ESBL rate was comparatively low (1.6 %), the percentages of ESBL-producing Enterobacter spp. and Escherichia coli increased from 0.8 and 0.5 %, respectively, ...

Uric acid is excreted by the kidneys and intestinal tract, while other routes are negligible. By addition of purines to a purine-free or low purine diet changes in uric acid turnover and excretion occur, which cannot be investigated by metabolic balance methods as that part of the uric acid turnover secreted into the gut is completely destroyed by the intestinal flora (Sorensen, 1960). We therefore studied uric acid pool size and turnover in normal subjects both during a purine-free diet and during ...
