Recently the numbers of spoilage incidents in food industry by the species of Thermoascus are increasing, but the risk of food spoilage have never been evaluated. It became obvious that their heat-resistances were higher than those of other heat-resist fungi, Byssochlamys, Hamigera and Neosartorya by our analyses. On the other hand, Thermoascus aurantiacus and Byssochlamys verrucosa had the idh gene, but they showed no patulin production in Potato dextrose broth or Czapek-glucose medium. Therefore, ...

In the beverage industry, peracetic acid has been increasingly used as a disinfectant for the filling machinery and environment due to merits of leaving no residue, it is safe for humans, and its antiseptic effect against fungi and endospores of bacteria. Recently, Chaetomium globosum and Chaetomium funicola were reported resistant to peracetic acid; however, little is known concerning the detail of peracetic acid resistance. Therefore, we assessed the peracetic acid resistance of the species of ...

Species of the genus Byssochlamys, heat-resistant fungi, cause the spoilage and poisoning of heat-processed acidic foods due to the formation of heat-resistant ascospores and the biosynthesis of patulin. Here we evaluated the heat resistance and patulin production of four species of Byssochlamys. Byssochlamys fulva was significantly more heat-resistant than the other species, and Byssochlamys lagunculariae and Byssochlamys nivea synthesized patulin. This is the first report of patulin production ...

Phospholipase A2 activity in lysates of mast cells such as rat mastocytoma RBL-2H3 cells and mouse bone marrow-derived IL-3-dependent mast cells (BMMC) was measured using phosphatidylcholine (PC), phosphatidylethanolamine (PE), or phosphatidylserine (PS) as a substrate. Both types of cells exhibited phospholipase A2 activity with a similar pH profile; the optimum pH observed with PS as a substrate was 5.5–7.4, whereas that with PE or PC was 8.0–9.0. PE and PC bearing an arachidonate at the sn-2 position ...

Species of the genus Neosartorya are heat-resistant fungi that cause the spoilage of heat-processed acidic foods due to the formation of heat-resistant ascospores, and they produce mycotoxins, such as fumitremorgins and gliotoxin. Their anamorphs are phylogenetically and morphologically very close to Aspergillus fumigatus, which has never been reported as a spoilage agent in heat-processed food products. Therefore it is important to discriminate between the species of Neosartorya and A. fumigatus ...

Human fibroblast growth factor 19 (FGF19) is an enterohepatic hormone that is involved in the regulation of hepatic metabolism of bile acids, lipids, and glucose. Farnesoid X receptor (Fxr)-null mice exhibit steatosis-like symptoms, showing higher hepatic lipid levels than with the wild-type mice. We investigated the influence of FGF19 treatment on hepatic lipogenesis in Fxr-null mice. Recombinant FGF19 treatment (400 μg/kg/d) for 3 d prevented the accumulation of lipid droplets and decreased serum ...

Abstract Self-directed behaviours (SDBs) are said to be indicative of negative emotions. The present study focused on chimpanzees’ SDBs during cognitive experiments in order to investigate how each chimpanzee reacted to his or her errors and to changes in task difficulty. We recorded and analysed the behaviour of six chimpanzees during cognitive experiments at the Primate Research Institute, Kyoto University, in Japan. We compared the rate of SDBs after correct trials versus incorrect trials, and ...

Phosphorylation of endogenous inhibitor proteins for type-1 Ser/Thr phosphatase (PP1) provides a mechanism for reciprocal coordination of kinase and phosphatase activities. A myosin phosphatase inhibitor protein CPI-17 is phosphorylated at Thr38 through G-protein-mediated signals, resulting in a >1000-fold increase in inhibitory potency. We show here the solution NMR structure of phospho-T38-CPI-17 with rmsd of 0.36 ± 0.06 Å for the backbone secondary structure, which reveals how phosphorylation triggers ...

Recent large trials on sodium–glucose cotransporter 2 (SGLT2) inhibitors showed that these agents improved cardiovascular outcomes in patients with type 2 diabetes (T2D) with a high risk of cardiovascular events (1). However, the underlying mechanisms of these clinical benefits and vascular effect of SGLT2 inhibitors in that patient group remain uncertain. Endothelial dysfunction is exacerbated by metabolic disorders, such as diabetes, and involved in the pathophysiology of diabetes-related cardiovascular ...

African apes and humans share a genetic mutation that enables them to effectively metabolize ethanol. However, voluntary ethanol consumption in this evolutionary radiation is documented only in modern humans. Here, we report evidence of the long-term and recurrent ingestion of ethanol from the raffia palm (Raphia hookeri, Arecaceae) by wild chimpanzees (Pan troglodytes verus) at Bossou in Guinea, West Africa, from 1995 to 2012. Chimpanzees at Bossou ingest this alcoholic beverage, often in large quantities, ...

Paraquat causes lung injury by oxidative stress. After 48 h of intraperitoneal administration of paraquat (50 mg/kg of body weight) to rats, the vitamin C concentration in the lungs was significantly decreased, while lung vitamin E content was increased after 12 h. These results indicate that vitamin C directly reflected the oxidative stress in the lungs.

Abstract Stem cell factor (SCF)/c-kit system is critical for human mast cell development. We thus examined the effects of STI571, an inhibitor of the c-kit tyrosine kinase receptor, on the proliferation and function of human mast cells. STI571 at concentrations of 10−6 M or higher almost completely abolished the SCF-dependent progeny generation from cord blood-derived cultured mast cells through an inhibition of the tyrosine phosphorylation of c-kit. The compound also suppressed the early phase of ...

Promyelocytic leukaemia zinc finger (PLZF) is a transcriptional repressor that was originally isolated from a patient with promyelocytic leukaemia. PLZF also affects key elements for cell cycle progression, such as cyclin A, and can affect the tumourigenicity of various cancers. Thus far, the behaviour of PLZF in thyroid carcinoma remains unclear. We analysed the expression profile of PLZF in different types of benign and malignant thyroid lesions as well as in normal thyroid tissue. Specifically, ...

Volumes of agglutinates produced by the microtiter latex antiglobulin test (LAGT) were measured by an electronic data processing (EDP) method using a particle volume distribution analyzer. This method proved highly sensitive for quantitating weak antibodies, such as anti-digoxin and anti-DNP. EDP measurement of the microtiter latex antiglobulin-inhibition test (LAGIT) is also a suitable method for quantitating HCG, digoxin and DNP.

Abstract The color development due to the complex formation of poly(vinyl alcohol) (PVA) with iodine increased with increasing syndiotacticity of PVA. Isotactic PVA showed no color development. The color development of syndiotacticity‐rich PVA film decreased with increasing annealing temperature for films before complexization, whereas that of atactic (commercial) PVA increased with it. Lower temperatures, the elongation of complex film, and the presence of boric acid enhanced the absorbance at 600 ...

Cyclophilins facilitate the peptidyl‐prolyl isomerization of a trans ‐isomer to a cis ‐isomer in the refolding process of unfolded proteins to recover the natural folding state with cis ‐proline conformation. To date, only short peptides with a cis ‐form proline have been observed in complexes of human and Escherichia coli proteins of cyclophilin A, which is present in cytoplasm. The crystal structures analyzed in this study show two complexes in which peptides having a trans ‐form proline, i.e. succinyl‐ ...

Induced pluripotent stem cells (iPSCs) are potentially valuable cell sources for disease models and future therapeutic applications; however, inefficient generation and the presence of integrated transgenes remain as problems limiting their current use. Here, we developed a new Sendai virus vector, TS12KOS, which has improved efficiency, does not integrate into the cellular DNA, and can be easily eliminated. TS12KOS carries KLF4, OCT3/4, and SOX2 in a single vector and can easily generate iPSCs from ...
