Life expectancy in the developed world is increasing, but this comes with a simultaneous explosion in 'age-related' as well as 'lifestyle-related' diseases, resulting in a decline in quality of life. Three such diseases are Type 2 diabetes mellitus (T2DM), Polycystic Ovarian Syndrome (PCOS) and non-alcoholic fatty liver disease (NAFLD), which all share a common reduced cellular response to the hormone insulin (termed insulin resistance). In T2DM, insulin resistance is clearly a contributing factor to ...

The insulin-stimulated phosphoinositide 3-kinase (PI 3-kinase)/3-phosphoinositide-dependent kinase-1 (PDK1)/protein kinase B (PKB) kinase cascade is believed to play a critical role in metabolic control and cell survival, largely mediated through PKB phosphorylation of many proteins. Recent findings demonstrate that the transcription factors FKHR (forkhead in rhabdomyosarcoma), AFX (ALL1 fused gene from chromosome X) and FKHRL1 (FKHR-like 1; termed FKHR isoforms) are phosphorylated by PKB in cells, leading ...

Desert pavement plays a fundamental role in arid land processes. The first step towards quantitatively analysing this role is detailed surficial mapping. To this end, map units were developed to represent the physical variability of a desert pavement surface at a large scale (decimeters to meters). Six distinct map units were defined by surface textural parameters of clast size, percent cover, and sorting through fieldwork on a basalt flow in the Mojave Desert. These surface mosaics abut with visually ...

Irrigation in semiarid regions can alter soil C sequestration processes compared with those of native soils. To better understand the effect of these altered processes, we studied the C sequestration and dynamics of two soils from major irrigated agricultural regions of California (the San Joaquin Valley and the Imperial Valley). Soils were sampled from selected native and cultivated fields to represent a span of almost a century of irrigated farming. Field soil samples were analyzed for total soil ...

Developing realistic soil carbon (C) sequestration strategies for China's sustainable agriculture relies on accurate estimates of the amount, retention and turnover rates of C stored in paddy soils. Available C estimates to date are predominantly for the tilled and flood-irrigated surface topsoil (ca. 30 cm). Such estimates cannot be used to extrapolate to soil depths of 100 cm since soil organic carbon (SOC) generally shows a sharp decrease with depth. In this research, composite soil samples were ...

Background: Chronic suppurative otitis media (CSOM) affects many children in disadvantaged populations. The most appropriate topical antibiotic treatment in children with persistent disease is unclear. Methods: Children with CSOM despite standard topical treatment were randomized to 6–8 weeks of topical ciprofloxacin (CIP) versus topical framycetin-gramicidin-dexamethasone (FGD). Otoscopic, audiologic, and microbiologic outcomes were measured using standardized assessments and blinding. Results: Ninety- ...

The LKB1 tumour suppressor phosphorylates and activates AMPK (AMP-activated protein kinase) when cellular energy levels are low, thereby suppressing growth through multiple pathways, including inhibiting the mTORC1 (mammalian target of rapamycin complex 1) kinase that is activated in the majority of human cancers. Blood glucose-lowering Type 2 diabetes drugs also induce LKB1 to activate AMPK, indicating that these compounds could be used to suppress growth of tumour cells. In the present study, we ...

Abnormal expression of the hepatic gluconeogenic genes (glucose-6-phosphatase [G6Pase] and PEPCK) contributes to hyperglycemia. These genes are repressed by insulin, but this process is defective in diabetic subjects. Protein kinase B (PKB) is implicated in this action of insulin. An inhibitor of PKB, Akt inhibitor (Akti)-1/2, was recently reported; however, the specificity and efficacy against insulin-induced PKB was not reported. Our aim was to characterize the specificity and efficacy of Akti-1/2 ...

ALG-2 (apoptosis linked gene 2 product) is a calcium binding protein for which no clear cellular function has been established. In this study we identified Scotin as a novel ALG-2 target protein containing 6 PXY and 4 PYP repeats, earlier identified in the ALG-2 binding regions of AIP1/ALIX and TSG101, respectively. An in vitro synthesized C-terminal fragment of Scotin bound specifically to immobilized recombinant ALG-2 and tagged ALG-2 and Scotin were shown by immunoprecipitation to interact in ...

ABSTRACT Many regions of southern California's coastal sage scrub (CSS) are rapidly declining as exotic annual plants replace native shrubs. During this conversion, the subsurface hydrology of the semiarid hillslopes that support CSS may be altered. This could chronically suppress the ability of native shrubland to revegetate the landscape since ecosystem processes of nutrient availability and of seedling establishment rely on spatial patterns of available soil water. In this work, soil water and ...

In 1991, the National Water Quality Assessment Program was initiated. The U.S. Geological Survey (USGS) as the lead agency worked with other federal, state, and local agencies to understand the spatial and temporal occurrence of water quality degradation, including the effects of both human activities and natural environmental factors. The results of this initiative were published as, “The Quality of Our Nation's Waters” (USGS, 1999), with specific reference to the influence of agricultural nutrients ...
