This chapter attempts to critically review the electrophysiological methods that have been used in the past to quantify epithelial shunt conductance. Emphasis of this chapter is on flat sheet epithelia. A number of specific problems involved with shunt analysis in tubular epithelia has been discussed in methodological reviews and are mentioned only briefly in this chapter. The methods to be discussed differ in their ability to represent the electrical properties of given epithelia appropriately. All are based on equivalent circuit representations of epithelia, which, for various reasons, are gross over simplifications. It yields accurate data only when the ionophore and the cell membrane have the same relative selectivities or respective permeabilities for all permeable ions (this includes, of course, membranes that are permeable for only one ion species in case they are investigated with an ionophore that is highly selective for this ion).
