ResumoThis chapter discusses the large-scale alkaline lysis method, which is used for purifying plasmid DNA from E. coli suitable for most molecular biological applications. It has an important advantage over the Triton–Lysozyme method in that it does not require ultracentrifugation as a step in purification and requires considerably less time. The single disadvantage of this method is that a small amount of E. coli chromosomal DNA and RNA is present in the final product, which complicates precise quantitation of plasmid yield using optical density measurements. The plasma purification method requires growing in advance, a 250-ml culture of E. coli containing the plasmid.
