ResumoThis chapter presents a procedure for the preparation and assay of substrates for β-glucuronidase. There are various requirements of a good substrate for assaying β-glucuronidase: (1) the glucuronide or a suitable derivative should be a crystalline product which can be efficiently purified, (2) it should be readily hydrolyzed by the enzyme, (3) the amount of aglucurone liberated should be accurately measured in microgram amounts, (4) the rate of hydrolysis should be linear during the course of the incubation, (5) interference by tissue constituents and by unhydrolyzed substrate should be negligible, and (6) the assay procedure should be convenient and rapid, requiring only the usual laboratory apparatus. The substrate phenolphthalein glucuronic acid satisfies these requirements. Its use in the assay of β-glucuronidase is now widespread and the results may be accepted with confidence.
