Resumomg/kg TID or placebo BID.On day O, each animal was equipped with a 14Fr.percutaneous esophagostomy cannula.The animals were then administered the designated drug over 3 days.On the fourth day, 8-channel radial manometry of the LES was performed in unsedated animals positioned in right lateral recumbency.Mean LES pressure, average maximal (Max) LES pressure and LES length were measured.A one week washout period was then followed by randomization to one of the other treatments and the process repeated.Results.Mean and Max LES pressures were significantly increased after treatment with both Celecoxih and Cisapride.The response to Cisapride and Celecoxib were comparable.No statistical difference in the LES length was found between treatment groups.Conclusio~ In the present study, we investigated the modifications of the canine LES under the inluence of Celecoxib, a COX-2 inhibitor.There was a significant increase in the mean and maximal LES pressures with both Celecoxib and Cisapride.We conclude that Celecoxib has a positive effect on the canine LES and that the effect is similar to that observed with Cisapride.
