This paper describes the formation of a silanization compound with nitrocellulose to form a thin film by spin coating on glass slide which can be used as a protein chip. This nitrocellulose film is adhered to the glass slide by spin coating directly and can immobilize protein by hydrophobic physical adsorption and epoxy chemical binding simultaneously. Using this spin coating thin film formation technique, both the Cy3 and Cy5 channel backgrounds are reduced by 90% when compared with the original FAST™ slide. The result of an alpha-1-foetoprotein (AFP) antigen sandwich experiment shows an S/N ratio of 68.8 in concentration of 10 ng/mL of AFP antigen, and the dynamic range is between 30 and 3000 ng/mL.
