Über den polysaccharidgehalt der serumeiweiss-fraktionen bei gesunden und kranken

Resumo1. Mit Hilfe der Eiweiβ- und Kohlenhydratfärbung nach papierelektrophoretischer Fraktionierung des Serums von Gesunden und Kranken sowie der Bestimmung der proteingebundenen Hexosen und der Mucoproteide dieser Seren, wird, neben dem rel. % gehalt der einzelnen Fraktionen an Eiweiβen und Kohlenhydraten der Polysaccharidgehalt von 100 g Eiweiβ jeder Fraktion ermittelt, der Anteil der Mucoproteide an der α1-Globulinfraktion und der Quotient von neutralen: sauren Kohlenhydraten dieser Fraktion bestimmt. 2. Die Albumin-, α1- und β-Globulinfraktion besitzen normalerweise durchschnittlich jeweils etwa 20%, die α2-Globulinfraktion etwa 27% und die γ-Globulinfraktion rund 12% der eiweiβgebundenen Polysaccharide. Die Standardabweichung ist gröβer als bei den Eiweiβfraktionen. Das Albumineiweiβ enthält rund 1 g%, das α1-Globulineiweiβ rund 12 g% Kohlenhydrate, von denen 31% den Kohlenhydraten der Mucoproteide angehören. Der Polysaccharidgehalt des α2-Globulineiweiβes beträgt rund 11 g%, der des β-Globulineiweiβes rund 5 g% und in 100 g γ-Globulineiweiβ sind rund 2 g Kohlenhydrate. 3. Neben entsprechenden Mittelwerten für Patienten mit epidemischer Hepatitis, chron. Hepatitis, Lebercirrhose, Cholecystitis, Carcinom verschiedener Lokalisation, Diabetes mellitus, generalisiertem Morbus Hodgkin, werden Einzelwerte für Fälle mit Lungentuberkulose, β-Plasmocytom und Lupus, Untersuchungsergebnisse mit zeitlichem Intervall bei Patienten mit Lebercirrhose mit und ohne Komplikation sowie bei einem β-Plasmocytom mitgeteilt und eine Unterteilung der Carcinompatienten aufgrund dieser Ergebnisse in Übereinstimmung mit dem klinischen Status in drei Gruppen vorgenommen. Die diagnostische, differentialdiagnostische und prognostische Bedeutung wird gestreift. 4. Die Leber wird als Bildungsstätte der möglicherweise für den Zellaufbau erforderlichen Mucoproteide angesehen. Mucoproteide wurden im Liquor, Ascites, Pleuraempyempunktat und in Leukocyten gefunden. Über Bildungsstätte und Funktion der proteingebundenen Hexosen kann keine Aussage gemacht werden. In zwei Fällen von Durchblutungsstörungen werden im arteriellen Blut weniger proteingebundene Hexosen gemessen als im venösen. 5. Der Kohlenhydratgehalt der neutralen Glykoproteide in der α1-Globulinfraktion ist bei Leberkrankheiten und Ulcus duodeni oder ventriculi gröβer, bei den anderen untersuchten Krankheiten niedriger. 6. Die besondere Bedeutung des Kohlenhydratgehaltes der γ-Globulinfraktion wird kurz diskutiert. 1. By means of protein- and carbohydrate-staining following paper electrophoretic fractionation of serum from healthy and sick persons, as well as by the determination of the protein-linked hexoses and mucoproteins of these sera, both the relative % content of protein and carbohydrate in the single fractions and the polysaccharide content of 100 g protein from each fraction, have been ascertained In addition, the mucoprotein portion in the α1-globulin fraction and the quotient of neutral: acid carbohydrate of this fraction have been determined. 2. Appropriate average values are given for patients with epidemic hepatitis, chronic hepatitis, cirrhosis of the liver, cholecystitis, carcinoma of various parts, Diabetes mellitus, and disseminated Hodgkin's disease, together with single values for cases with tuberculosis of the lungs, β-plamocytoma and lupus, and also examination results obtained at intervals on patients with cirrhosis of the liver with and without complications, and in one case of β-plasmocytoma. Carcinoma patients have been sub-divided into 3 groups on the basis of these results in conformity with the clinical condition. The diagnostic, differential diagnostic and prognostic importance is touched on. 4. The liver is important as the site of formation of the mucoproteins, which are possibly necessary for the building up of the cell. Mucoproteins have been found in cerebrospinal fluid, in ascites, in the liquid obtained by thoracic empyema puncture, and in leucocytes. No statement can be made about the site of formation and function of the protein-bound hexoses. In two cases of impaired blood supply less proteinbound hexoses were found in arterial blood than in venous blood. 5. The carbohydrate content of the neutral glycoproteids in the α1-globulin fraction is greater in liver diseases and ulcus duodeni or ventriculi, and lower in other diseases investigated. 6. The particular significance of the carbohydrate content of the γ-globulin fraction is briefly discussed. 1. Les auteurs donnent des valeurs moyennes pour des patients souffrant d'hépa- tite épidémique, d'hépatite chronique, de cirrhose du foie, de cholécystite, de carcinomes de diverses localisations, de diabète sucré, de la maladie de Hodgkin généralisée; ils indiquent de plus des valeurs isolées pour des cas de tuberculose pulmonaire, de plasmocytome β et de lupus, des résultats d'expériences faites avec un certain intervalle de temps chez des patients souffrants de cirrhose du foie avec et sans complications de même que chez un plasmocytome β. En se basant sur ces résultats et en accord avec l'état clinique, ils établissent une division des patients souffrants de carcinomes en trois groupes. La signification pour la diagnose, la diagnose différen- tielle et la prognose est mentionnée en passant. 4. Le foie est considéré comme endroit de formation des mucoprotéides qui sont peut-être nécessaires pour la formation des cellules. L'on a trouvé des mucoprotéides dans le liquor, les ascites, le liquide d'une pleurésie purulante et les leucocytes.
