ResumoDoses of quinidine gluconate ranging from 15 to 50 mg/kg were dripped slowly iv into pentobarbital anesthetized openchest dogs over a 3 hr period. Control dogs received only saline. Hemodynamic measurements were recorded during the infusion period. The animals were then sacrificed, and left ventricular tissues were taken immediately for electron microscopy. No ultrastructural changes were observed in the control or 15 mg/kg dogs, but damage to mitochondria of the cardiac cells was noted increasingly at the larger doses of 30 and 50 mg/kg. The mitochondrial involvement was primarily one of cristolysis, i.e., the dissolution of the internal membranous folds. At the highest dose an increase in the length and size of some mitochondria was seen. Other cellular organelles were not structurally altered by the drug.
