Effect of peritubular infusion of angiotensin II on rat proximal nephron function

Resumo

Effect of peritubular infusion of angiotensin II on rat proximal nephron function. The effect of infusing Ileu 5 -angiotensin II into the peritubular capillaries on the rate of proximal fluid reabsorption (RPFR) was studied using sodium pentobarbitalanesthetized rats. In each rat fluid collections were made alternately from different tubules associated with peritubular infusion of either Ileu 5 -angiotensin II or isotonic saline solution. Infusion rate was approximately 20 nl·min −1 throughout. RPFR was derived from timed quantitative collections of tubular fluid from the last superficial proximal segments. Each fluid sample was collected at the free-flow pressure (FFP) measured during peritubular infusion. FFP's measured prior to infusion were similar in the two groups of tubules. FFP's increased by 6.0cm of H 2 O with the infusion of 20 ng·ml −1 of angiotensin II, by 1.1cm of H 2 O with 0.1 ng·ml −1 of angiotensin II and only by 0.3cm of H 2 O with isotonic saline infusion. Significant reductions in RPFR (Δ = -7.4 nl·min −1 ·g of kidney wt −1 ) and TF/P inulin (Δ = -0.79) were found in tubules associated with 20 ng·ml −1 of angiotensin II infusate. Nephon GFR was similar in the two groups of samples (Δ = 0.7 nl·min −1 ·g of kidney wt −1 ). When the infusate was 0.1 ng·ml −1 of angiotensin II, the values for RPFR and nephron GFR were similar to those obtained from tubules associated with saline infusion. Tubular fluid samples were collected alternately from different tubules with isotonic saline infusion or without peritubular infusion. Nephron GFR and RPFR were almost the same value in these two groups of samples. The average hydrostatic presure in the first order peritubular capillaries was 23.3cm of H 2 O with 20 ng·ml −1 of angiotensin II infusate; 17.1cm of H 2 O with 0.1 ng·ml −1 of angiotensin II infusate; 17.0cm of H 2 O with saline infusate and 16.6cm of H 2 O without peritubular infusion. The results indicate that the presence of Ileu 5 -angiotensin II on the capillary side of a renal tubule may cause a significant reduction in the rate of tubular fluid reabsorption in the proximal convoluted tubule. Effet de l'infusion peritubulaire d'angiotensine II sur la fonction du tube proximal chez le rat. L'effet de l'infusion de Ileu 5 -angiotensine II dans les capillaires peritubulaires sur le RPFR (debit de reabsorption proximale) a ete etudie chez des rats anesthesies par le pentobarbital. Des collections ont ete faites, pour chaque rat, dans des tubules dont les capillaires ont ete infuses soit avec Ileu 5 -angiotensine II soit avec du solute sale isotonique. Le debit d'infusion etait approximativement de 20 nl.min −1 . RPFR a ete calcule a partir de collections minutees et quantitatives du fluide tubulaire dans la derniere anse superficielle. Chaque echantillon tubulaire a ete obtenu a la FFP (pression en debit libre) mesuree au cours de l'infusion peritubulaire. Les FFP mesurees avant l'infusion etaient similaires dans les deux groupes de tubules. Les FFP ont augmente de 6,0cm H 2 O avec un infusat a 20 ng.ml −1 d'angiotensine II, de 1,1cm H 2 O avec un perfusat a 0,1 ng.ml −1 d'angiotensine II et seulement de 0,3cm H 2 O au cours de l'infusion de solute sale isotonique. Des diminutions significatives de RPFR (Δ = -7,4 nl.min −1 .g KW −1 ) et de TF/P inuline (Δ = -0,79) ont ete observees dans les tubules avec une infusion de 20 ng.ml −1 d'angiotensine II. Le debit de filtration glomerulaire du nephron etait semblable dans les deux groupes d'echantillons (Δ = 0,7 nl.min −1 .g KW −1 ). Quand l'infusat etait a 0,1 ng.nl −1 en angiotensine II, les valeurs de RPFR et du debit de filtration du nephron (NGFR) etaient semblables a celles obtenues a partir de tubules correspondants a une infusion de solute sale. Des echantillons tubulaires ont ete collectes alternativement a partir de tubules correspondants a une infusion de solute sale ou sans infusion peritubulaire. NGFR et RPFR ont practiquement les memes valeurs dans ces deux groupes d'echantillons. La moyenne des pressions hydrostatiques dans les capillaires peritubulaires du permier ordre etait de 23,3cms H 2 O avec 20 ng.ml −1 d'angiotensine II; 17,1cms H 2 O avec 0,1 ng.ml −1 ; 17,0cms H 2 O avec un infusat de solute sale et 16,6cms H 2 O sans infusion peritubulaire. Ces resultats indiquent que la presence d'Ileu 5 -angiotensine sur le versant capillaire d'un tube renal peut determiner une diminution significative de la reabsorption tubulaire dans le tube contourne proximal.
