Resumoα-Sarcin is a cytotoxin produced by a mold; it inhibits protein synthesis by inactivating ribosomes. The toxin selects and cleaves a single phosphodiester bond near the 3′ end of the large RNA ribosomal subunit, whereas it hydrolyses naked RNA at purines in both single- and double-stranded regions. These remarkable properties have made α-sarcin valuable in analysing the structure of ribosomes and in determining the binding sites for proteins in ribonucleoprotein complexes.
