ResumoThree major pathways to apoptosis-associated caspase activation have been identified in mammals: the extrinsic or death receptor pathway, the intrinsic or apoptosome pathway and the cytotoxic lymphocyte-initiated granzyme B pathway.1 The order of events in each of these pathways to caspase activation is by now relatively well understood. However, recent studies including one published in this issue of CDD2, have generated new insights into caspase activation pathways in response to signals that provoke cell damage, as well as in contexts in which caspase activation does not result in apoptosis.
