ResumoThe erect of ouabain, a specific sodium-potassium dependent adenosine triphosphata-se (Na+-K+-BTPase) inhibitor, on antigen-induced histamine release was studied using guinea pig lung fragments sensitized in vitro with rabbit antibodies against bovine serum albumin.Histamine was assayed spe&ofEuorometrically.When sensitized tissue had been preincubated with ouabain (< 1.0 x lo-' M) for 10 min prior to antigenic challenge, release of histamine wm sign$ieantly inhibited (maximum 54q& p < 0.001, N = 9, paired t test).The most significant inhibition was obtained near the optimal concentration of antigen.The inhibition was dependent on the length of preinoubation (5 60 min), and was partially reversible upon washing the tissu.eremoving the ouabain.Ouabain did not seem to prolong the duration of the histamine release process.Increase in potassium ion (5 1.1 x 10-j Y) inhibited the histamine release and had additive erects to ouabain notion.Dibutyryl cyclic dMP (5 5 x 10-j H), which aould enhance the release, strongly antagonized the inhibition.Glucose removal from the medium did not abolish the ouabain efect.The results seem to indicate that immunologic release of histamine is under the influence of the membrane Na+-K+d TPase activity.Recent reports indicate that phosphatidylserine enhances antigen-induced release of histamine from sensitized rat mast cells.1-4Since phosphatidylserine seems to stimulate or be required for the Nat-K+-ATPase of various cells in vertebrates,5-g the enhancement of antigen-induced histamine release by this compound might be related to the activation of the ATPase.In this study, we used a specific antagonist of Na+-K+-ATPase, and observed inhibition of the antigen-induced histamine release by this antagonist.
