ResumoAbstractWe present a new direct method to study the binding capacity of a hormone conjugate to cells in histological preparations. The methodused an HCG preparation labeled with 1 nm colloidal gold, which was visualized for light microscopy by silver enhancement. In an indirect control, a monoclonal antibeta HCG antibody was used to detect the bound HCG in a two step irnrnunogold-silver staining method with the antimouse antibody labeled with colloidal gold. The results showed that positive staining reactions in a variety of tissues are highly dependent upon the presence of HCG in the incubation procedure. These positive reactions may be due to the binding of HCG to the receptor. This method could open new ways to study hormone/cell binding capacities and possible interactions.
