Resumo

Lipid modifications of proteins can be detected by incubation of cultured cells with radioactive precursors followed by detergent extraction of protein and analysis of the extract by SDS-PAGE and fluorography. Identification of particular proteins requires immunoprecipitation, affinity purification, or differential expression (such as overexpression) before SDS-PAGE analysis. The purpose of this article is to present a brief description of protein acylation and prenylation modifications that have been observed and the details of how to detect the modifications by metabolic radiolabeling of cellular proteins.
