ResumoThe reaction of imidazole and imidazole derivatives with formaldehyde can be demonstrated with NMR techniques. The results show that only one nitrogen of the imidazole ring reacts to form a N-hydroxymethyl derivative in alkaline solution. Under acidic conditions both nitrogen positions can support N-hydroxymethyl derivatives. This reaction represents a useful tool for the further investigation of enzyme mechanisms involving the imidazole nucleus.
