ResumoThree acid phosphatase isozymes have been isolated from human erythrocytes by chromatography on DEAE-Sephadex. Complete resolution of the isozymes was obtained. The individual nature of the isozymes was confirmed by the differences in their pH optima, response to activation and inhibition, and electrophoretic mobility. The isozymes are electrophoretically indistinguishable from the three isozymes identified by electrophoresis of crude hemolyzates. The isolation procedure is readily adapted to the preparation of larger quantities of the isozymes. Variations in the characteristics of previously isolated and purified erythrocyte acid phosphatases are interpreted with respect to these three isozymes.
