Abstract The isolation and analysis of DNA from parchment has the potential to provide a great deal of information about the material that was difficult or impossible to obtain before. We review progress in recovering DNA from other ancient and historic materials, and the limited amount of work done so far with parchment and other manuscript material. We discuss the likelihood of being able to recover well preserved DNA from parchment, and the questions that could in principle be addressed. Finally, we comment on the technical advances needed for the field to develop significantly and reliably, and implications for conservators. Résumé «Les possibilités d'extraction et d'exploitation de l'ADN du parchemin: un examen des possibilités et des obstacles» L'isolement et l'analyse de l'ADN du parchemin peut potentiellement fournir un grand nombre d'informations sur la matière qui étaient difficiles ou impossibles à obtenir auparavant. Nous passons en revue les progrès accomplis dans la récupération de l'ADN d'autres matériaux anciens et historiques, et l'avancement des travaux réalisés, jusque là limités, avec le parchemin et les autres matériaux manuscrits. Nous discutons de la probabilité d'être en mesure de récupérer un ADN bien conservé du parchemin, et les questions qui pourraient en principe être abordées. Enfin, nous commentons les progrès techniques nécessaires dans ce domaine qui restent à développer significativement et efficacement, et nous commentons les implications pour les restaurateurs. Wir untersuchen sowohl die Fortschritte, die die Entnahme von DNA aus anderen antiken oder historischen Materialien gemacht hat, als auch die begrenzten Untersuchungen, die bisher mit Pergament und anderen Manuskriptmaterialien stattgefunden haben. Wir diskutieren auch die Wahrscheinlichkeit, gut erhaltene DNA aus Pergament zu gewinnen und die Fragen, die daraus im Prinzip beantwortet werden könnten. Schließlich kommentieren wir die technischen Fortschritte, die nötig sind, um diese Feld zuverlässig und stark zu entwickeln und was für Konsequenzen dies für Konservatoren und Restauratoren hat.
