Resumo

Some of the electrophoretic parameters are examined with respect to their effect on restricting the amount of RNA analyzable per gel. Acrylamide concentration was found to be the most important. By addition of a dilute pregel to the analyzing gel, up to 0.8–1 mg RNA can be examined with excellent resolution on gels with diameter 6 mm, and up to 2.5 mg RNA on gels with diameter 10 mm. The suggested principle might also be used to increase the yield of preparative electrophoresis. The large amounts of RNA introduce special difficulties in the scanning of the ultraviolet absorption. Suggestions are made on how to overcome these difficulties.
