ResumoSummary A new method of isolating potato protoplasts from cultured roots is described. Roots of in vitro grown plants are cut into small fragments and cultured in a defined medium to induce branching. Subsequently protoplasts are isolated from root apexes, which are the only tissues susceptible to enzyme action. Protoplasts are cultured to produce calli, thereafter to the regeneration of whole plants. The morphological characterization and cultural behaviour of the protoplasts are discussed and related to their tissue origin.
