ResumoThree methods of isolation of nuclei of HeLa cells were investigated in connection with studies of pulse-labeled RNA in HeLa cells: (a) sucrose-homogenization, (b) osmotic shock, (c)_citric acid. Of the three methods only the nuclei isolated by citric acid method conserved all the radioactivity of the pulse-labeled RNA found in the nuclei of whole cells. Isolation by the other two methods resulted in a 20 and 17% loss of the labeled RNA of the nuclei.
