ResumoThe Side Effects of Drugs Annuals form a series of volumes in which the adverse effects of drugs and adverse reactions to them are surveyed. The series supplements the contents of Meyler's Side Effects of Drugs: the International Encyclopedia of Adverse Drug Reactions and Interactions. This literature review is mainly composed of a special review on adverse reactions to methotrexate. A major part of the review is a description of the many drug–drug interactions that have been described with methotrexate, including interactions with acetylsalicylic acid, alcohol, aminoglycoside antibiotics (paromomycin and neomycin), aminophenazone (amidopyrine), amiodarone, benzimidazoles, bleomycin, bosentan, carbamazepine, carboxypeptidase G2, cephalosporins, chloral hydrate, choline magnesium trisalicylate, ciclosporin, ciprofloxacin, cisplatin, co-trimoxazole (trimethoprim + sulfamethoxazole), etanercept, etoposide, etretinate, fluorouracil, glucocorticoids, ibuprofen, ion exchange polymers (colestimide and colestyramine), nitrous oxide, non-steroidal anti-inflammatory drugs (NSAIDs; azapropazone, diclofenac, etodolac, flurbiprofen, ibuprofen, indometacin, ketoprofen, loxoprofen, lumiracoxib, naproxen, phenylbutazone, piroxicam, and rofecoxib), metamizole (dipyrone, noramidopyrine), penicillins, phenobarbital, phenytoin, pristinamycin, probenecid, proton pump inhibitors, sulfasalazine, tacrolimus, tetracyclines, theophylline, thiazide diuretics, triamterene, trimethoprim, and vaccines. The management of methotrexate toxicity using alkalinization, folic acid and folinic acid, and aminophylline is also briefly discussed. The review also refers to literature published in 2011 on bleomycin, oxaliplatin, and paclitaxel.
