Isolation of acetylpeptide from the protein of cucumber virus 4

ResumoFrom the peptic digest of the protein of cucumber virus 4, three ninhydrin-negative, acidic peptides, Ala[Tyr,AspNH2], [Ala,Tyr,Asp,Pro]Ileu.Thr and [Ala,Tyr,Asp]Pro. Ileu.Thr.Ser, were isolated by the aid of an ion exchange column. An additional acidic peptide was obtained from the chymotryptic digest of the protein and its amino acid sequence was determined as N-acetyl.Ala.Tyr. The structure of this acetyldipeptide was confirmed by comparison with synthetic material. Combining the structure of these four peptides it is possible to give the following structure for the longest, a heptapeptide: Acetyl.Ala.Tyr.AspNH2.Pro.Ileu.Thr.Ser. The acetylpeptides are probably derived from the N-terminal end of the polypeptide chain since no N-terminal group has ever been found in the cucumber virus 4 protein.
