In vitro transformation of protoplast-derived Hyoscyamus muticus cells by Agrobacterium tumefaciens

Protoplasts of Hyoscyamus muticus were treated with Agrobacterium tumefaciens strain C58 in vitro and three transformed cell lines were established. These cell lines initially produced nopaline, although one lost this ability after six months in culture. A highly increased frequency of hormone independent growth was observed after incubation with A. tumefaciens strain LBA4058. No transformants, however, could be detected. The results demonstrate that the in vitro transformation with A. tumefaciens is not restricted to Nicotiana tabacum.
