ResumoUsing glutaraldehyde as a coupling reagent, human IgG was conjugated with horseradish peroxidase. Rabbits were hyperimmunized against human IgG. Pieces of spleen were then incubated with the peroxidase-conjugated IgG and the localization of the antigen-antibody reaction was revealed cytochemically with the substrate of peroxidase. This reaction is confined to the plasma cell line and is observed in the ergastoplasm, perinuclear space, Golgi apparatus and in ergastoplasm-bound ribosomes. Such a method permits the electron microscopiacl demonstration of antibodies at the ribosomal level.
