Resumo

A new activator is reported for coupling phosphoramidites to a free 5'-hydroxyl group during oligonucleotide synthesis. Pyridinium trifluoroacetate/N-methyl imidazole is a remarkably efficient replacement for 1H-tetrazole in the solid-supported synthesis of oligonucleotides. This reagent is safe and inexpensive, is not moisture-sensitive, and is soluble in acetonitrile.
