ResumoHyphae of Penicillium chrysogenum Thom have an extension zone which is about 1.5 μm long. At the base of the extension zone about 5% of the cytoplasmic volume is composed of vesicles with a mean diam of 68 nm. The concentration of these vesicles increases throughout the extension zone, until finally they occupy about 45% of the cytoplasmic volume at the extreme tip of the hypha. A Spitzenkörper containing microvesicles with a mean diam of 29 nm is also present at the hyphal tip. Wall rigidification at the base of the extension zone involves the formation of a substantial secondary wall. The effects of pH, temperature and nutrient limitations on wall thickness of chemostat cultures of P. chrysogenum are described.
